摘  要:目的观察黄芪甲苷对体外培养胚胎神经干细胞增殖作用的影响,并探讨其作用机理。方法体外分离培养大鼠胚胎神经干细胞,Nestin免疫细胞化学染色进行神经干细胞的鉴定,BrdU标记鉴定增殖。采用稀释法检测黄芪甲苷高、中、低剂量组(6、0.6、0.06μmol/L)对体外培养胚胎神经干细胞增殖作用的影响。结果 Nestin染色为阳性,BrdU的增殖鉴定为阳性。与对照组相比,黄芪甲苷高、中、低剂量组神经球生成数目明显增加。结论黄芪甲苷能够明显增加神经球生成数目,具有促进胚胎神经干细胞体外增殖的作用。Objective To investigate the effect of astragaloside on the proliferation of neural stem cells(NSCs) in vitro.Methods The primitive NSCs of embryonic day 16 SD rat were cultured in vito.The NSCs were identified by immunocytochemical(ICC) staining of Nestin.The ICC staining of BrdU was adopted to characterize proliferation of NSCs.The effect of the different dosage astragaloside groups(6,0.6, 0.06 μmol/L) on proliferation of NSCs was valued by limited dilution method.Results The NSCs were successfully cultured in vitro.Characteristic protein(Nestin) of NSCs labeling and BrdU labeling were positive.Compared with the control group,the numbers of neruospheres of the different dosage astragaloside groups were increased obviously.Conclusion Astragaloside can significantly increase the number of neruospheres and promote the proliferation of NSCs in vitro.
