摘 要:目的利用生物转化方法来生产具有抗氧化和抗肿瘤活性药物羟基化去氢表雄酮。方法利用中空海藻酸钙胶囊对白色链霉菌进行固定化,然后用此固定化菌对甾体类药物去氢表雄酮(DHEA)进行生物转化,并对固定化菌的转化条件进行优化,以期获得高产量的羟基化DHEA。结果中空海藻酸钙胶囊制备的固定化白色链霉菌对DHEA具有转化作用,并且可以通过固定化白色链霉菌的二次转化来实现羟基化DHEA产率的提高,我们同时获得了固定化白色链霉菌的最优转化条件:对生长两天的白色链霉菌进行固定化,温度35℃,pH值7.0,转速150r/min,转化时间3d。结论采用固定化白色链霉菌转化DHEA方法可行,而且此种方法具有广泛的工业应用价值。Objective Producing hydroxylated DHEA with antioxidant and anticancer activity by bioconversion methods. Methods Through the hollow calcium alginate to immobilize Streptomyces albus to transform DHEA. And through the optimization of the immobilization and transformation conditions as well as the second transformation of the immobilized bacteria to get the high production of hydroxylated DHEA. Results The immobilized bacteria had the transformation activity on DHEA, and we got the high production of hydroxylated DHEA by the second transformation of immobilized bacteria. The best transformation conditions of immobilized bacteria was:immobilized the Streptomyces albus which had grown for two days,temperature 35℃ , pH 7,rotati on speed 150r/min,transformation time 3days. Conclusion It is possible for immobilized bacteria to transform DHEA,and this method has wide industrial application value.
