摘  要:目的探讨+Gz暴露后大鼠脑皮层自噬相关标志物Beclin-1和LC3表达的变化。方法雄性SD大鼠30只,随机分为对照组、+10 Gz/3 min暴露组和+10 Gz/5 min暴露组。+Gz暴露组分别于暴露后6 h、24 h取脑皮层组织,采用RT-PCR和Western Blot方法分别测定自噬相关基因Beclin-1和LC3 mR-NA及蛋白表达的变化。结果与对照组相比,两组+Gz暴露后6 h脑皮层Beclin-1和LC3 mRNA及蛋白表达均显著增强(P<0.01),暴露后24 h其表达量仍显著增加(P<0.05)。+10 Gz/5 min组较+10Gz/3 min组表达增加更为明显(P<0.05)。结论 +10 Gz暴露可诱导脑皮层自噬相关标志物Beclin-1和LC3的高表达,提示高+Gz应激促进了脑皮层自噬的发生,且随着暴露时间的延长,自噬表达更显著。Objective To investigate the expression of autophagy related markers, Beclin-1 and LC3, in rat cer- ebral cortex after + Gz exposure. Methods A total of 30 male SD rats were randomly assigned to control, + 10 Gz/3 min and + 10 Gz/5 rain groups. The cerebral cortex tissues of rats subjected to + 10 Gz were taken at 6 h and 24 h after exposure, respectively. RT-PCR and Western Blot were applied to determine the expression of autophagy related genes, Beclin-1 and LC3. Results Compared with the control group, the expression of Beclin-1 and LC3 in the cerebral cortex were significantly increased after 6 h exposure in both two + Gz groups (P 〈0.01), and remained significantly higher after 24 h exposure (P 〈0.05). The expression of Beclin-1 and LC3 were significantly higher in + 10 Gz/5 min group compared with + 10 Gz/3 rain group (P 〈0.05 ). Conclusion + 10 Gz exposure increases the expression of autophagy related genes, Beclin-1 and LC3, sugges- ting the induction of autophagy by + Gz stimulus in the cerebral cortex in rats. Furthermore, the induction of autophagy is amplified with the extension of exposure time under the same level of + Gz stimulus.
