右旋柠烯对人结肠癌LS174T细胞p-ERK和p-Akts表达的影响

摘　　要：目的研究右旋柠烯（D—limonene）对人结肠癌细胞（LSl74T）中信号传导分子ERKl／2、Akt及其磷酸化蛋白（P—ERK和P—Akts蛋白〉表达的影响。方法采用四甲基偶氮唑盐（MTF）方法检测右旋柠烯对LSl74T细胞增殖的抑制作用；采用Westernblot方法检测不同浓度右旋柠烯对LSl74T细胞中ERKl／2、Akt及其磷酸化蛋白表达情况的影响。结果右旋柠烯作用LSl74T细胞48h后，呈剂量依赖性抑制细胞增殖；Westernblot结果显示右旋柠烯对细胞中ERKl／2、Akt蛋白的表达没有明显影响，但显著下调P—ERKl／2蛋白和P—Akts的表达。结论右旋柠烯抑制LSl74T细胞增殖可能与其抑制P—ERKl／2和P—Akts表达有关。Objective To investigate the effect of D - limonene on the expressions of p - ERK and p - AKTs in human colon cancer cell line LS174T. Methods The effect of D - limonene on the cell proliferation was measured by methyl thiazolyl tetrazolium （MTI＇）, and the expression of ERK1/2, Akt and their phosphorylated proteins （ p - ERK and p - AKTs） were detected by Western blot. Results D - limonene inhibited the prolifera- tion of LSI74T cell in a dose dependent manner,because the LS174T cell were exposed to D -limonene for 48 hours. D - limonene did not influence the expressions of ERK1/2 and Akt, but it significantly decreased p - ERK1/2 and p - Akts expressions. Conclution It is suggested that D - limonene could inhibit proliferation of LS174T cell via inhibiting the p - ERK1/2 and p - Akts pathway.
