摘 要:目的比较不同剂量东莨菪碱对小鼠行为和学习记忆功能的影响,确定东莨菪碱致小鼠记忆障碍模型的合适剂量。方法 48只昆明种小鼠,随机分为4组,每组12只。对照组腹腔注射等体积0.9%氯化钠溶液,其他各组分别腹腔注射东莨菪碱2,3,5 mg·kg-1。回避反应法检测行为和学习记忆能力,分光光度法测定大脑皮质和海马区乙酰胆碱(ACh)、乙酰胆碱酯酶(AChE)及乙酰胆碱转移酶(ChAT)含量及活力。结果东莨菪碱3,5 mg·kg-1组小鼠上、下台逃避潜伏期显著缩短(P<0.05),小鼠脑ACh、AChE和ChAT含量和活力受到显著影响。结论东莨菪碱对小鼠行为和记忆障碍损伤的程度随剂量增大而增强,3,5 mg·kg-1均能显著造成小鼠学习记忆行为损伤,显著影响小鼠大脑皮质和海马区Ach的合成与代谢;东莨菪碱对小鼠的行为及记忆功能损伤的合适剂量为3 mg·kg-1。Objective To compare the effects of different doses of scopolamine on behavior, learning and memory function in mice and determine the appropriate dose of scopolamine for inducing memory impairment of mice model. Methods The total of 48 kunming mice were randomly divided into 4 groups (control group and three model groups, each containing 12 mice) were given with scopolamine by intraperitoneal injection at the doses of 2,3 and 5 mg·kg^-1 ,and the same volume of saline, respectively. Learning and memory ability were investigated by avoidance reaction and the content and vitality of Ach, AChE and ChAT were determined by a spectrophotometric method. Results ① The latency period of scopolamine at 3 mg·kg^-1 dose and 5 mg·kg^-1 dose was significantly reduced (P〈0.05) ;②The vitality and content of ACHE, ChAT and Ach in the brain of these mice were remarkably affected. Conclusion ①The degree of damage of scopolamine to the behavior and memory of mice is increased with the dose ;learning and memory behaviors in mice were significantly injury by 3 mg·kg^-1 dose and 5 mg·kg^-1 of mice ② Scopolamine can significantly impact acetylcholine synthesis and metabolism in the cerebral cortex and hippocampus of mice at the doses of 3 mg·kg^-1 and 5 mg·kg^-1 ;③The appropriate dose of scopolamine on the behavior and memory function injury of mice is 3 mg·kg^-1.
