摘  要:目的研究奥沙利铂长循环脂质体的处方筛选和制备工艺,并研究其稳定性。方法采用乙醇注入法制备奥沙利铂长循环脂质体,以包封率为评价指标,采用正交设计优化处方和制备工艺,高效液相色谱法测定脂质体包封率。结果最佳工艺处方为卵磷脂与胆固醇质量比5∶1,奥沙利铂10 mg,聚乙二醇2000浓度0.05%,平均包封率(51.87±0.90)%。结论该制备方法较简单,经优化制备的奥沙利铂长循环脂质体处方合理,工艺可行,稳定性好。Objective To optimize technology on preparation of oxaliplatin long-circulating liposome. Methods The liposome was prepared by ethanol injection method. Orthogonal design was used to optimize the prescription and preparation. Encapsulate rate of the liposome was determined by high performance liquid chromatography. Results The optimum prescription was as follows: the quality ratio of lecithin to cholesterol was 5 : 1, the amount of oxaliplatin was 10 mg, and the concentration of polyethylene glycol 2000 was 0.05% , the average coating rate is (51.87± 0.90 )%. Conclusion The optimized formulation of PEG2000-conjugated oxaliplatin long-circulating liposome is reasonable, oracticable and stable.
