摘 要:根据禽源隐孢子虫GP20基因,设计1对保守引物和1对特异性引物,建立巢氏PCR检测方法,并进行验证.该巢氏PCR能特异性地扩增出496bp的禽源隐孢子虫基因组DNA中相应片段,最低能检测到0.52fg的阳性参照DNA.建立的巢氏PCR方法具有高度的特异性和敏感性,可以用于样品的检测.通过该方法鉴定山东省德州地区养鸡场中感染鸡的隐孢子虫种类为C.baileyi,感染率为31.5%.To develop a nested PCR assay for detection of chicken--derived Cryptosporidium, two pairs of primers were designed based on the bird--derived GP20 sequences. These primers selectively amplified a 496bp DNA fragment gene of bird --derived Cryptosporidium. With these primers, a nested PCR assay was developed. The results showed that the target fragment was present in DNA extracts of fecal samples from chicken infected with Cryptosporidium, The assay was able to detect as low as 0.52 fg of positive control DNA. This study showed that PCR was a highly sensitive and specific method and could serveas a valuable tool for detection of Cryptosporidium. the Cryptosporidium isolates were C. baileyi by this meth- od and the infection rate was 31.5% in Dezhou.
