5-FU变形脂质体制备及局部治疗兔喉气管狭窄的研究

摘　　要：目的评价5-氟尿嘧啶(5-FU)变形脂质体对兔瘢痕性喉气管狭窄的治疗效果。方法利用逆向蒸发法制备5-FU变形脂质体,并检测其形态、包封率等特性。采用刮擦法制备喉气管狭窄模型,环形刮除气管黏膜约周径2/3,诱导气管内瘢痕形成,将诱导瘢痕性喉气管狭窄成功的动物模型随机分为3组:5-FU变形脂质体组(A组)、5-FU水溶液组(B组)以及生理盐水组(C组),各组分别在内镜监视下经皮穿刺将药物注射入瘢痕内部。分别在用药后第10、20、30天纤维喉镜下观察气管狭窄情况。结果治疗初期5-FU变形脂质体组和水溶液组无差别,30天5-FU变形脂质体组狭窄度显著低于水溶液组。结论 5-FU变形脂质体对兔瘢痕性喉气管狭窄的治疗有一定的疗效,可以作为喉气管狭窄治疗方法进一步研究。Objective To evaluate the efficacy of deformable liposomes encapsulated with 5 - FU in treatment of laryngotracheal ste- nosis in rabbit models. Methods The 5 - FU deformable liposomes was prepared by the reverse - phase evaporation method, and the a- morphous size distribution and the encapsulation efficiency of it was investigated. The tracheal mucosa were scraped about 2/3 of weeks di- ameter with a nylon brush to induce the scar in airway grow, then models were divided into three groups：5 - FU deformable liposomes group,5 - FU group and saline group, in which drug was injected into scar of every group by paracentesis guided under endoscope respec- tively. The stenosis degree was observed under laryngofiberoscope 10,20,30 days after administrated. Results Airway stenosis of 5 - FU deformable liposomes group had no significant difference as compared with 5 - FU group at 10 days after administration, but 5 - FU de- formable liposomes significantly reduced the airway stenosis at 30 days after administration as compared with 5 - FU group again. Conclu- sion Deformable liposomes encapsulated with 5 - FU was effective for laryngotracheal stenosis which suggests that it has petential as a new method for ameliorating airway stenosis originated from granulation tissue.
