摘 要:目的:通过观察电针传统针刺助产处方(合谷-三阴交穴)对孕晚期大鼠子宫平滑肌协调收缩的影响,进而部分阐释传统针灸助产处方的科学内涵。方法:选用健康成年初孕Wistar雌鼠22只,随机分为电针治疗组和妊娠模型对照组;健康成年未孕Wistar雌鼠11只做为空白对照组。电针治疗组孕鼠均在妊娠第20天上午进行电针,以先电针双侧合谷穴20min,再加刺双侧三阴交穴5min,共针刺25min,电流强度0.2-0.3mA,2/50Hz疏密波为干预方法。同一时间段,模型对照组与空白对照组大鼠布袋束缚25min。采用酶联免疫吸附实验(ELISA)检测大鼠血清中雌二醇(E2)、孕酮(P)及子宫组织中前列腺素F2α(PGF2α)的含量,并采用蛋白免疫印记(Western Blotting)法检测大鼠子宫组织中缝隙连接蛋白-43(Cx-43)的表达。结果:与空白对照组比较,模型对照组和电针治疗组血清E2、P、E2/P含量均显著升高(P<0.05,P<0.01),子宫PGF2α含量和Cx-43表达均显著升高(P<0.01);与模型对照组比较,电针治疗组血清E2含量显著升高(P<0.05),P含量下降但无显著差异,E2/P升高但无显著差异,子宫PGF2α含量和Cx-43表达显著升高(P<0.05)。结论:电针刺激优化传统针灸助产处方通过改变孕晚期大鼠血清及子宫组织中相关激素水平而促进孕晚期大鼠子宫向活跃期转变,并通过增加子宫缝隙连接蛋白的表达而发挥其促进子宫协调收缩的作用。可认为是该处方发挥促分娩作用的主要途径之一。部分阐释传统针灸助产处方的分子生物学机制,为针灸在产科中的应用提供科学依据。Objectives: To study the effect of electro-acupuncture (EA) at a classical inducing-labor acupuncture formula [Hegu (LI4)- Sanyinjiao (SP6)] on uterine smooth muscle coordinated contraction in late-stage pregnancy rat, so as to partly explore its scientific mechanism of inducing labor. Methods: Twenty-two healthy in early-stage pregnancy Wistar rats were randomly divided into a model control group and an EA group. And eleven healthy adult female Wistar unpregnancy rats were included in a normal control group. In the morning of the 20th day of pregnancy, the EA group was applied EA at bilateral Hegu (LI4) for 20min plus Sanyinjiao (SP6) 5min, a total of 25min was required. The intensity of EA was 0.2-0.3mA with 2/50Hz sparse-dense wave. Rats in control groups were tied 25rain with bags at the same time. Outcome measures including Estradiol (E2), Progesterone (P) in serum and Prostaglandin F2a (PGF2a) in tissue were detected by Enzyme Linked Immunosorbent Assay (ELISA). And expression of Connexin 43 (Cx-43) was measured by protein immunoblotting (Western Blotting). Results: Compared with the normal control group, the contents of E2, P and E2/P in serum, PGF2a and Cx-43 expression in the uterus in the model control group and the EA group were significantly increased (P〈0.05, P〈0.01). Compared with model control group, the contents of E2 in serum, PGF2a and the Cx-43 expression in the uterus in the EA group were significantly increased (P〈0.05), the EA group showed decreased serum P and increased serum EJP, but with insignificant differences. Conclusion: Classical inducing-labor acupuncture formula combined with optimized EA stimulation parameters could active the uterus of late-stage pregnant rats by regulating the serum and uterus hormone levels, promote uterine coordinated contraction by increasing the expression of gap junction protein in smooth muscle of uterus.
