摘  要:目的:探讨精制逍遥散治疗抑郁症的作用机制。方法:将40只健康SD雄性大鼠随机分为正常组、模型组、逍遥散组和氟西汀组,后3组进行大鼠抑郁症造模。逍遥散组、氟西汀组于实验的第1天开始分别灌服精制逍遥散、氟西汀,每日1次。21d后观察大鼠行为学以及海马区P450scc蛋白表达的变化。结果:抑郁症模型大鼠体质量增长缓慢,糖水消耗率降低,摄食潜伏期延长,5min内移动的速度减慢,5min内移动总距离、进入中央区停留时间减少;海马CA3和DG区P450scc蛋白表达明显减弱(P<0.05,P<0.01)。逍遥散组及氟西汀组体质量、糖水消耗率、摄食潜伏期、5min内移动速度及总距离和中央区停留时间与模型组比较有统计学意义,CA1区相对增加,并能有效逆转抑郁症大鼠海马CA3和DG区P450scc蛋白表达。结论:大鼠抑郁模型脑区存在神经活性甾体的功能紊乱,逍遥散治疗抑郁症其机制可能是通过调节P450scc蛋白表达。进一步影响神经活性甾体的功能而实现的。Objective: To explore the mechanism of refined Xiaoyao Powder for treatment of depression. Methods: Forty healthy SD male rats were randomLy divided into a normal group, a model group, a Xiaoyao Powder group and a Fluoxetine group. The depression rat model was established in the latter three groups. From the first day of the experiment, the Xiaoyao Powder group was orally administrated the refined Xiaoyao Powoler once every day; the rats in the Fluoxetine group were treated with Fluoxetine once every day. After treatment of 21 days, changes of ethology and expression of protein P450scc in hippocarnpus were observed in the rats. Results: In the depression model rats, the body mass inereased slowly, sucrose preference decreased and ingestion latency stage extended, movement speed within 5rain reduced, and total motion distance within 5rain, stay period in center zone decreased; the expression of P450scc in the CA3 and DG regions of hippocampus significantly reduced (P〈0.05, P〈0.01). In the Xiaoyao Powder group, and the Fluoxetin group the body mass, sucrose preference, ingestion latency stage, movement speed and total motion distance within 5min, stay period in center zone had statistical differences compared with the model group, relatively increased the expression in CA1 region and could effectively reverse the expressions of P450scc in hippocampus CA3 and DG regiens. Conclusion: The depression model rat has dysfunction of the neurosteroids and Xiaoyao Powder could regulate its functions. The mechanism is possibly carded out by regulating the functions of P450scc.
