摘 要:目的:观察中枢及外周5-羟色胺(5-HT)在抑郁症肝郁脾虚证大鼠中的变化规律,探索抑郁症肝郁脾虚证的生物学基础,为证候的客观化诊断提供参考依据。方法:采用慢性不可预知温和应激(CUMS)建立大鼠抑郁症模型,通过肝郁脾虚证临床四诊信息等效转化为大鼠宏观表征的方法来进行大鼠的证候判别。Elisa和免疫组化法测定大鼠中枢和外周5-HT含量。结果:应激6-8周后抑郁症大鼠出现稳定的肝郁脾虚证。与正常组相比,CUMS组大鼠海马5-HT含量持续降低(P<0.05,P<0.01),血清和小肠组织中5-HT含量持续升高(P<0.05,P<0.01)。结论:肝郁脾虚证与中枢5-HT的持续降低及外周血液和小肠组织中5-HT的持续升高相关。Objective: To observe the variation regularity of central and peripheral 5-HT on depressive syndrome model of stagnation of liver and deficiency of spleen in rats, to explore the biological foundation of depressive syndrome of stagnation of liver and deficiency of spleen and provide a reference basis for clinical diagnosis. Methods: Rat depression mode/was established by chronic unpredictable mild stress (CUMS). The syndrome model of stagnation of liver and deficiency of spleen was selected from the depression model by equivalently transforming the related clinical symptoms for on of rats. Elisa and cal method were used to measure the content of central and peripheral 5-HT. Results: The stable syndrome time period was supposed to be 6-8 weeks after stress. In the CUMS group, compared with the normal control group, the content of 5-HT in the hippocampus area had a sustained decrease (P〈0.05, P〈0.01) while the content of 5-HT in serum and small intestine tissue increased steadily (P〈0.05, P〈0.01). Conclusion: The syndrome of stagnation of liver and deficiency of spleen is associated with the sustained decrease of central 5-HT and increase of peripheral 5-HT in serum and small intestine tissue.
