针灸内关穴预处理对兔缺血再灌注损伤心肌细胞死亡受体通路Fas/FasL蛋白表达的影响

摘　　要：目的:探讨针灸内关穴预处理对兔缺血再灌注损伤心肌细胞死亡受体通路Fas/FasL蛋白表达的影响。方法:将24只家兔随机分为假手术组、缺血再灌注组、电针组和艾灸组。采用结扎左冠状动脉前降支40min再灌注60min的方法,建立心肌缺血再灌注损伤模型。电针组和艾灸组分别在针/灸预处理之后48h再行缺血再灌注。应用免疫组织化学法,以累积光密度为主要指标观察Fas/FasL蛋白表达的水平。结果:缺血再灌注组心肌组织中Fas蛋白的阳性表达较假手术组增多(P<0.05),电针组和艾灸组较缺血再灌注组均显著降低(P<0.05),且两组间无显著性差异;缺血再灌注组和艾灸组心肌组织中FasL蛋白的阳性表达较假手术组增多(P<0.05),电针组较缺血再灌注组显著降低(P<0.05)。结论:针灸内关穴预处理能抑制死亡受体通路Fas/FasL蛋白的表达,从而抑制缺血再灌注后细胞凋亡的发生,由此对心肌细胞发挥延迟相的保护作用。Objective： To study the effect of preacupuncture ＇Neiguan（PC6）＇ on Fas/FasL protein expression in death receptor pathway of myocardial ischemia reperfusion injury cell in rabbits. Methods： 24 rabbits were randomly divided into sham- operation group, ischemia reperfusion group, electroacupuncture group and moxibustion group. The ischemia reperfusion models were established with ligation of the left anterior descending coronary artery lasting for 40min utes and followed by reperfusion for 60 minutes. The rats in electroacupuncture group and moxibustion group received the acupuncture of moxibustion treatment for 48h before ischemia reperfusion. Then the expression of Fas/FasL was measured by imrnunohistochemical technique, with integral optical density as the main index to observe the Fas/FasL protein expression. Results： The Fas expression was more significantly in the ischernia reperfusion group than the sham-operation group （P〈0.05）, but significantly decreased in telectroacupuncture group and moxibustion group （P〈0.05）, and there was no obvious difference between the last two groups. The expression of FasL markedly increased in the ischemia repeffusion and moxibustion groups with comparison to that in sham-operation group （P〈0.05）; and those in electroacupuncture group significantly decreased than in ischemia reperfusion group （P〈0.05）. Conclusion： Preacupuncture ＇Neiguan （PC6）＇ can inhibit the expression of Fas/FasL protein in death receptor pathway so that to inhibit the apoptosis after ischemia reperfusion, thus expressing delayed protection for myocardial cells.
