摘 要:目的:观察左归丸、右归丸对老年大鼠学习记忆相关信号分子脑源性神经营养因子(BDNF)、酪氨酸蛋白激酶B(TrkB)、细胞外调节蛋白激酶(ERK1/2)、反应结合蛋白(CREB)、活性调节细胞骨架蛋白(Arc)及突触蛋白(Synapsin)mRNA表达的影响。方法:以自然衰老SD雄性大鼠为动物模型,随机分为老年对照组、老年糖皮质激素拮抗组、老年左归组和老年右归组,另设青年对照组。采用原位杂交法检测海马各分区TrkB mRNA表达变化,实时定量PCR法观察海马组织BDNF、ERK1/2、CREB、Arc及Synapsin mRNA表达变化。结果:与青年对照组比较,老年大鼠海马上述分子mRNA表达均明显降低(P<0.05);与老年对照组比较,RU38486能明显上调老年大鼠海马CA1区TrkB mRNA表达,提高海马组织ERK1/2、Arc及Synapsin mRNA表达(P<0.05);左归丸及右归丸均能明显上调老年大鼠海马CA1、CA2-3分区TrkB mRNA表达(P<0.05),同时左归丸还能显著提高老年大鼠海马组织Arc及Synapsin mRNA表达(P<0.05)。3种药物对其余信号分子的表达均存在一定的上调作用,但无统计学差异。结论:左归丸、右归丸及RU38486可能通过改善学习记忆相关信号分子的异常表达,进而提高长时程增强效应和延缓老年大鼠学习记忆能力的退化。Objective: To observe the effects of kidney-tonifying recipe-Zuogni Wan and Yougui Wan on the expressions of learning and memory related signal molecules (BDNF, TrkB, ERK1/2, CREB, Arc and Synapsin mRNA). Methods: As senile animal model, the nature old male SD rats wbre randomly divided into four groups: senile group, senile and antagonist RU38486, senile and Zuogui Wan group, senile and Yuogui Wan group, and there uas young group in addition. The expression of TrkB mRNA was examined by in situ hybridization method. The expressions of BDNF, ERKI/2, CREB, Arc and Synapsin were tested by Real-time quantitative PCR. The regulating effects of kidney-tonifying recipe on above indices were also studied. Results: Compared with young group, the expressions of mRNA-BDNF, TrkB, ERK1/2, CREB, Arc and Synapsin in senile group were obviously decreased (P〈0.05). Compared with the senile group, Ru38486 could obviously raise TrkB mRNA expression in CA1 areas of hippocampus (P〈0.05). Zuogui Wan and Yougui Wan could raise TrkB mRNA expression in CA1 and CA2-3 areas of hippocampus (P〈0.05). And Yougui Wan could obviously raise Arc and Synapsin mRNA expressions of hippocampus as well (P〈0.05). Conclusion: The deterioration of learning and memory abilities in senile rats might be partly related to the abnormal transduction of the signal pathway which was mediated by BDNF. Kidney- tonifying recipe and RU38486 could improve spatial learning and memory abilities of senile rats by down regulating the higher CORT levels and retarding aging changes of the expressions of related signal transduction molecules.
