摘 要:目的:探讨新风胶囊(XFC)对佐剂关节炎(AA)大鼠肺功能及调节性T细胞(Treg)、Th细胞的影响。方法:将48只大鼠随机均分为正常对照(NC)、模型对照(MC)、雷公藤多苷片(TPT)、XFC组。除NC组外,其余各组大鼠右后足跖皮内注射弗氏完全佐剂0.1mL致炎,致炎后第19天给药。NC组及MC组给予0.9%氯化钠溶液,治疗组分别给予XFC 2.4g/kg、TPT 10mg/kg,每天1次,连续给药30d。观察各组大鼠足跖肿胀度、关节炎指数(AI),ELISA法检测血清细胞因子γ-干扰素(IFN-γ)、白介素-4(IL-4),流式细胞术检测外周血Treg,定量PCR及免疫印迹检测肺组织IFN-γ、IL-4、FoxP3表达。结果:与NC组比较,MC组大鼠足跖肿胀度、AI升高,肺功能降低,血清和肺组织IFN-γ、Th1/Th2升高,IL-4、FoxP3和外周血Treg降低(P<0.05,P<0.01)。与MC组比较,XFC组足跖肿胀度、AI降低,肺功能升高,血清和肺组织IFN-γ(mRNA和蛋白)降低,外周血Treg和IL-4、FoxP3(mRNA、蛋白)升高,且优于TPT组(P<0.05,P<0.01)。结论:新风胶囊通过调节Treg表达,促使Th1向Th2漂移,纠正Th1/Th2细胞失平衡状态,维持细胞免疫抑制状态,减少炎性介质对肺组织刺激,提高肺功能水平。Objective: To study the effect of Xinfeng Capsule (XFC) on pulmonary function and regulatory T cells (Treg), Th cells. Methods: Forty-eight rats were randomly divided into a control (NC) group, a model control (MC) group,a tripterygium glycosides tablet (TPT) group and a XFC group. In addition to the NC group, rats were induced to AA model by intradermally injecting 0.1mL Freund's complete adjuvant into the rear fight paw. After 19-day administration, the NC group and the MC group were given 0.9% sodium chloride solution, while the treatment groups were given XFC 2.4g/kg, TPT 10mg/kg, respectively, once a day, continuous administration for 30 days was required. The degree of paw swelling and arthritis index (AI) were observed. Gamma-interferon (IFN-γ), interleukin-4 0L-4) of serum were detected by ELISA, Treg of peripheral blood was detected by flow cytometry, and the level of IFN-γ, IL-4, FoxP3 of lung tissue were detected by quantitative PCR and Western Bloting. Results: Compared with the NC group, the degree of paw swelling, AI, IFN-γ, Thl/Th2 were increased, and the expressions of pulmonary function, IL-4, FoxP3, Treg were decreased in the MC group (P〈0.05, P〈0.01). Compared with the MC group, paw swelling and AI were reduced, while pulmonary function parameters, Treg, IFN-γ (mRNA and protein), IL-4, FoxP3 (mRNA and protein) were elevated in XFC group. It was superior to those in the TPT group (P〈0.05, P〈0.01). Conclusion: Xinfeng Capsule could adjust the level of Treg in peripheral blood, and prompt the drift that of Thl to Th2. It also could correct imbalance of Thlfrh2, maintain cellular immune suppression, reduce inflammatory mediators in lung tissue stimulation to improve pulmonary function.
