摘  要:目的:探讨糖肾宁对链脲佐菌素(STZ)诱导糖尿病肾病(DN)大鼠的肾脏及其TGF-β1/Smad7信号转导通路的影响。方法:Wistar大鼠54只,二次随机分为正常组10只,模型组15只、厄贝沙坦组15只和糖肾宁组14只。2周测大鼠体质量;每4周收集大鼠24h尿液,考马斯亮蓝G-250法测24h尿蛋白定量。14周时处死大鼠,检测血清尿素氮(BUN)、血清肌酐(Scr)、甘油三脂(TG)、一氧化氮(NO)、丙二醛(MDA)、超氧化物歧化酶(SOD);大鼠肾组织行HE、Mallory染色并行病理学评分;免疫组化、原位杂交检测肾组织TGF-β1、Smad7、NF-κB蛋白及其mRNA表达。结果:糖肾宁可以改善DN大鼠的一般状况,升高DN大鼠体质量及降低肾质量体质量比(P<0.01),明显降低24h尿蛋白定量(P<0.01),减轻肾组织病理性损害。与模型组比较,糖肾宁组BUN、Scr、TG、MDA含量显著降低(P<0.01),NO、SOD含量明显升高(P<0.01);肾组织TGF-β1、NF-κB蛋白及mRNA表达明显降低(P<0.01),Smad7蛋白及mRNA表达显著升高(P<0.01)。结论:糖肾宁可能通过调控TGF-β1/Smad7信号转导通路,减轻肾脏病理损害而保护肾功能,进而延缓其病程进展。Objective: To discuss the renal protective effect of Tang Shen Ning capsule on diabetic nephropathy rats (DN rats) induced by streptozotocin (STZ) and the TGF-β1/Smad7 signal transduction pathway. Methods: 54 Wistar rats were randomly divided into the follow groups for twice: the normal control group (n=10), the model group (n=15), the irbesartan group (n=15), and the Tang Shen Ning group (n=14). The body weight at an interval of 2 weeks; 24 hours urine by collecting every four- week intervals was detected its protein content by coomassie brilliant blue G-250 method. Rats were executed on the 14th week. BUN, Scr, TG, MDA, NO and SOD in the blood serum were tested; histopathological scores and the HE, Mallory staining ways were applied to the kidney tissues. Via immunohistochemistry and in situ hybridization, the renal protein of TGF-β1, Smad7, NF- kB and their mRNA expressions were detected. Results: The general condition of rats could be improved by Tang Shen Ning capsules; the weight of DN rats could be increased and the kidney weight/body weight (KW/BW) could be reduced (P〈0.01); the 24 hours urinary protein were significantly reduced(P〈0.01); pathological damage of kidney could be relieved. Compared with the model group, the contents of BUN, Scr, TG, MDA could be obviously reduced (P〈0.01), and NO, SOD increased apparently (P〈0.01); the expressions of TGF-β1, NF-kB and their mRNA were significantly reduced (P〈0.01), while Smad7 and its mRNA could be increased (P〈0.01). Conclusion: It was found that Tang Shen Ning capsule had the function of protecting kidney, through regulating the expression of the TGF-β1/Smad7 signal transduction pathway. And it could put off the progress of DN rats.
