银屑灵优化方对银屑病豚鼠及炎性刺激角质形成细胞增殖的影响

摘　　要：目的:观测银屑灵优化方对豚鼠银屑病模型和角质形成细胞(HaCaT)增殖的影响。方法:心得安(盐酸普萘洛尔)乳剂涂抹豚鼠耳部造成银屑病模型,观察银屑灵优化方对该模型的病理状态影响并用Baker法评分;建立脂多糖(LPS)诱导的HaCaT细胞增殖模型,用MTT法检测不同浓度和时间下银屑灵优化方对HaCaT细胞的抑制率。结果:银屑灵优化方低、高剂量(5.98、11.96g/kg)能够显著降低银屑病豚鼠的Baker评分(P<0.01);浓度为250、125、62.5μg/mL银屑灵优化方对共同培养48h的HaCaT细胞有不同程度抑制作用(P<0.01),各个浓度(250、125、62.5、31.25、15.6μg/mL)银屑灵优化方对培养72h的HaCaT细胞均有抑制作用,差异有统计学意义(P<0.01)。结论:银屑灵优化方能够改善豚鼠银屑病病变,抑制HaCaT细胞过度增殖。Objective： The present study is to determine the anti-psoriatic effects of optimized Yinxieling Formula （OYF） on psoriasis-like model of guinea pigs and on lipopolysaccharide （LPS）-stimulated HaCaT keratinocyte cell line. Methods： Propranolol hydrochloride-induced psoriasis-like model in guinea pigs were applied to investigate the effects of OYF in vivo, and pathological morphology assessment was examined with Baker score. Anti-proliferative effects of OYF were examined in LPS-stimulated HaCaT keratinocyte cell line by MTT. Results： Administration of OYF in low or high dose（5.98, ll.96g/kg） decreased the Baker scores and ameliorated the psoriasis-like model of guinea pigs （P〈0.01）. OYF significantly inhibited the proliferation of LPS-stimulated HaCaT keratinocyte ceils in dose of 250, 125, 62.51ag/mL after co-cultured for 48h（P〈0.01）. When co-cultured for 72h, dose of 250, 125, 62.5, 31.3, 15.61ag/mL had inhibitive activity as well（P〈0.01）. Conclusion： The results indicated that the anti-psoriatic effects of OYF involved down-regulation psoriatic indexes in animal model and inhibition of proliferation in HaCaT keratinocyte cell line.
