摘 要:目的:探讨颐脑解郁方对缺氧/复氧条件下离体海马神经元突触素表达的影响。方法:以正常大鼠、经颐脑解郁方及尼莫地平灌胃大鼠血清为媒介,制备正常及缺氧/复氧损伤后海马组织星形胶质细胞条件培养液。提取乳鼠海马神经元并进行体外培养,根据所应用干预条件培养液的不同随机将神经元分为正常组、模型组、中药组、西药组4组。采用MTT法观察缺氧3h(is-3h)及复氧24h(rp-24h)时点海马神经元活性并计算细胞存活率,同时应用免疫细胞化学方法测定海马神经元突触素的表达。结果:is-3h时点模型组神经元活性、存活率及突触素表达均较正常组明显降低(P<0.01),与中药组比较无明显差异;rp-24h时点中药组上述指标均较模型组明显升高(P<0.01)。结论:颐脑解郁方可明显促进缺氧/复氧损伤后海马神经元突触素表达的增强,该方可能通过有效增加神经元突触密度而对缺血/再灌注损伤神经功能恢复发挥明显改善作用。Objective: To explore the effect of Yinao Jieyu prescription on the synaptophysin expression of the damaged hippocampal neurons induced by simulated hypoxia/reoxygenation in vitro. Methods: Four different astrocyte conditioned mediums were made based on the application of the intervention of the serums which contained different medicines. Rat hippocampal neurons were cultured and divided into 4 groups according to these different astrocyte conditioned mediums including normal group (N), model group (M), Chinese medicine group (CM) and western medicine group (WM). The activity and viability of the damaged neurons both at 3 hours after hypoxia and 24 hours after reoxygenatlon were evaluated by colorimetric MTT assay. The expression of synaptophysin in hippocampal neurons was measured by the method of immunocytochemistry. Results: The activity, viability and synaptophysin expression in M were all significantly lower than N (P〈0.01), and there was no difference in CM at 3 hours after hypoxia. And at 24 hours after reoxygenation, the activity, viability and synaptophysin expression in CM were much higher than M (P〈0.01). Conclusion: Yinao Jieyu prescription could improve the synaptophysin expression in hippocampal neurons with the hypoxia/reoxygenation injury. And Yinao Jieyu prescription might play an important role in the recovery of neurological function of the ischemia/reperfusion injury by effectively increasing the synaptic density.
