摘  要:为明确山东省番茄灰霉病菌的类型分布、遗传多样性以及遗传群组的划分与地理来源之间相关性。采用形态学观察结合5.8SrDNA-ITS序列分析法对单孢分离所得66个菌株进行了鉴定,利用ISSR标记技术对菌株进行遗传多样性分析,并采用UPGMA法进行聚类统计。鉴定结果显示,66个菌株均为灰葡萄孢菌(Botrytiscinerea),通过序列比对,该66个菌株的ITS序列属于2种类型,并且2种ITS序列只有单碱基差异。ISSR遗传多样性分析,结果获得24个ISSR分子标记,其中58.33%的片段具有多态性,UPGMA聚类分析将66个菌株划分为4个遗传聚类群。以上结果表明,菌株种类与遗传多样性均不丰富,遗传群组的划分与菌株的地理来源存在一定的相关性。In order to know the type distribution, the genetic diversity and the correlation between genetic group division and geographical origin of Botrytis cinerea in Shandong Province. In this paper, the morphological observation combined with 5.8S rDNA-ITS sequence analysis was used to identify the 66 isolations that obtained through the method of single spore, and the genetic diversity was analyzed by the inter-simple sequence repeats (ISSR), at the same time the cluster statistics was analyzed with UPGMA method. Appraisal results showed that 66 isolates were all the Botrytis cinerea. In addition, all the ITS sequences of 66 isolates were divided into 2 types through the sequence alignment, and only a single base differences between 2 types. Through ISSR genetic diversity analysis 24 sites were generated in total among which 58.33% were polymorphic. By UPGMA cluster analysis, the isolates were classified into 4 subgroups. All the above results indicated that strain types and kinds of genetic diversity were not rich, genetic group division and strain geographical source had certain relevance.
