摘  要:目的探讨南蛇藤提取物抑制人肝癌转移的分子机制。方法人肝癌HCCLM6细胞生长于培养液中进行传代,取对数生长期细胞进行实验。南蛇藤提取物组共分为10、20、40、80、160mg/L5个浓度,阳性对照组给予5-氟尿嘧啶(5-Fu)注射液180mg/L,阴性对照组不做处理。分别观察南蛇藤提取物对HCCLM6细胞增殖和迁移能力的影响,并检测南蛇藤提取物对HCCLM6细胞及其培养上清中血管内皮生长因子-C(VEGF-C)蛋白表达的影响。结果南蛇藤提取物组作用24、48h时160μg/ml对细胞增殖的抑制率最高(P<0.01)。南蛇藤提取物组和阳性对照组穿膜细胞数较阴性对照组明显减少(P<0.01);南蛇藤提取物组10、20、40、80μg/ml浓度穿膜细胞数多于阳性对照组,并呈浓度依赖性(P<0.05或P<0.01)。南蛇藤提取物组各浓度和阳性对照组VEGF-C蛋白水平均低于阴性对照组(P<0.05或P<0.01)。结论南蛇藤提取物能抑制HCCLM6细胞的增殖、迁移及下调VEGF-C蛋白的表达水平,这可能是其抑制肝癌转移的部分机制。Objective To study the molecular mechanism of Celastrus orbiculatus extract on liver cancer metastasis. Methods Human liver cancer HCLM6 cells were grown and passaged in the culture medium and the logarithmic growth phase cells were selected. The Celastrus orbiculatus extract group was divided into 10 mg/L, 20 mg/L, 40 mg/L, 80 mg/L and 160 mg/L concentrations. The positive control group was given 180mg/L 5-fluorouracil (5-FU) injection and the negative control group was not treated. The effects of Celastrus orbiculatus extract on the proliferation and migration of HCCLM6 cells were observed. The expression of vascular endothelial growth factor-C (VEGF-C) was observed as well. Results The inhibition rate of cell proliferation at 160 μg/ml was the highest after 24 hours and 48 hours of administration in the Celastrus orbiculatus extract group (P〈0.01). Comparing with the negative control group, the penetrated cells in the Celastrus orbiculatus extract group and positive control group were significantly decreased (P〈0.01). Comparing with the positive control group, the penetrated cells at 10μg/ml, 20μg/ml, 40μg/ml and 80μg/ml in the Celastrus orbiculatus extract group were significantly increased with a dose-dependent manner (P〈0.05 or P〈0.01). Comparing with the negative control group, the contents of VEGF-C protein were significantly decreased in the Celastrus orbiculatus extract group and positive control group (P〈0.05 or P〈0.01). Conclusion Celastrus orbiculatus extract can inhibit the proliferation and migration of HCCLM6 cells and downregulate the expression of VEGF-C, which might be part of the mechanism of celastrus orbiculatus extract on cancer metastasis.
