摘 要:目的观察重组日本血吸虫中国大陆株脂肪酸结合蛋白(rSj14FABP)免疫动物所诱导的免疫保护效果。方法用rSj14FABP分别对昆明系小鼠、Wistar大鼠及绵羊进行3次免疫,于第3次免疫后2周,分别用40、600、及300条尾蚴攻击,于攻击后6~8周剖杀小鼠和大鼠,11周剖杀绵羊,观察各实验组的减虫率及绵羊免疫组的组织、粪便减卵率。结果在小鼠试验中,以BCG为佐剂进行皮内免疫获得33. 7%的减虫率(P<0.05),明显高于以 FCA/FIA为佐剂进行肌肉免疫所获得的减虫率4.7%(P>0.05)。用rSj14FABP皮内免疫大鼠,获得的减虫率为31.9%,但和对照组相比无显著性差异。用rSj14FABP皮内免疫绵羊,减虫率为59.2%(P<0.01),同时,自攻击后第6周至11周,绵羊免疫组粪便中平均每克虫卵数(EPG)分别比对照组减少23.6%~63.3%,第11周解剖时,肝组织EPG减少44.9%,小肠EPG减少69.6%,大肠EPG未见减少。结论rSj14FABP能诱导抗日本血吸虫攻击感染的部分保护作用,值得作为抗日本血吸虫候选疫苗抗原进一步探讨。Objective To observe the protection induced by vaccination in animals with recombinant Schistosoma japonicum Chinese strain fatty acid binding protein (rSj14FABP). Methods Kunming mice, Wistar rats and sheep were vaccinated 3 times with rS14FABP and challenged with 40, 600 and 300 cercariae of Sj 2 weeks after final vaccination respectively. The mice and rats were perfused 6~8 weeks post-challenge and the sheep were perfused 11 weeks post-challeng. The reduction percentage of worm count in all vaccinated animals and egg count reductions in feces and tissues of sheep were calculated. Results in the experiment mice, the animals vaccinated I. D. with BCG as adjuvant got 33.7% worm reduction, significantly higher than that got in mice vaccinated I. M. with FCA/FIA as adjuvant (4.7%, worm reduction, P>0.05). In rats vaccinated I. D. without adjuvant, the worm reduction rate was 31.9% (P>0.05). In sheep vaccinated I. D. without adjuvant, the worm reduction rate was 59.2% (P<0.01), and the EPG reduction rates in feces were 23.6%~63.3% from week 6 to week 11 post-challenge. the EPG reduction rates in liver and small Intestine were 44.9% and 69.6% respectively comparing with the control animals, but no EPG reduction was seen in large intestine. Conclusion Vaccination with rSi14FABP could induce protection against S. japonicum infection. It is worthy to do more studies on this antigen as anti-schistoso miasis vaccine candidate.
