荧光光谱法研究左氧氟沙星和头孢呋辛与牛血清白蛋白分子的相互作用

摘　　要：在模拟动物生理条件(pH=7.40的Tris-HCl缓冲溶液中)下,左氧氟沙星(LVFX)和头孢呋辛(CEFX)能够猝灭牛血清白蛋白(BSA)的荧光。据此建立了利用荧光光谱法研究抗生素左氧氟沙星(LVFX)和头孢呋辛(CEFX)间相互作用的研究。结果表明:用Stern-Volmer方程处理实验数据发现BSA与药物发生反应生成了新的复合物,猝灭机理以静态猝灭为主,药物与蛋白结合位点数约为1,药物间存在相互作用,使药物与蛋白间的结合常数增大、结合稳定性增大,游离型药物含量减少。In the aqueous solution of Tris-HC1 （pH= 7. 40）, both levofloxacin （LVFX） and cefuroxime（CEFX） can quench the fluorescence of bovine serum albumin（BSA）. Based on this,study on the interaction between I.VFX and CEFX by fluorescence spectometry was established. The research showed that the quenching mechanism of the combination of bovine serum albumin and drug is a static procedure. The number of binding sites is about 1 in several system. It was proved that the interaction between the drugs increased the binding stability of the drug and protein.
