摘 要:目的观察Kruppel样因子4(KLF4)在创伤新生血管中的表达及在体外对内皮细胞功能的影响。方法用24只大鼠作皮肤创伤模型,分别于创伤后即刻、2、4、6、8、10、12、14d处死并取创缘组织作免疫组织化学染色;用KLF4小干扰RNA(siRNA)转染人脐静脉内皮细胞(HUVECs)48h后,检测其增殖、迁移和成管能力变化。结果KLF4在新生血管内皮细胞中有表达;转染KLF4 siRNA后,HUVECs的吸光度(A)值、迁移距离和成管环数明显降低,分别为0.948±0.043、(206.0±5.6)μm、(12.0±0.7)个(P〈0.05)。结论KLF4对皮肤创伤新生血管的形成起重要作用。Objective To investigate the expression of Kruppel-like factor4 (KLF4) in neovascular endothelial cells during the process of wound healing and the effect on endothelial cells function in vitro. Methods The skin wound model was established in 24 rats, and the marginal tissues were taken for im- munohistochemical staining after the rats were killed immediately, 2, 4, 6, 8, 10, 12, and 14 days after the trauma. Proliferation, migration and tube formation of human umbilical vein endothelial cells (HUVECs) were measured after transfection with KLF4 small interfering RNA (siRNA) for 48 h. Results KLF4 was expressed in neovascular endothelial ceils. The absorbance of HUVECs proliferation, the migration distance and tube number were 0. 948 ±0. 043, (206. 0±5.6) μm and 12. 0 ±0. 7 respectively (P 〈 0. 05) after transfection of KLF4 siRNA. Conclusion KLF4 plays an important role in the vascular formation (angiogenesis) during cutaneous wound healing.
