摘  要:目的观察钙离子抑制剂维拉帕米对周围神经损伤修复后瘢痕形成抑制作用。方法健康雄性SD大鼠60只,建立大鼠坐骨神经横断伤后端端吻合模型,右侧为实验组,左侧为对照组,将浸泡于200μmoL/L维拉帕米溶液至饱和的明胶海绵包裹在神经吻合口周围,左侧为对照组,将吸附生理盐水至饱和明胶海绵包裹在神经吻合口周围,6周后处死大鼠,取吻合口部位的神经组织做形态学检测,Western blot及免疫组织化学检测,称取双侧腓肠肌湿重并计算萎缩指数,行统计学分析。结果实验组肌肉湿重高于对照组(P〈0.05),肌肉萎缩指数实验组低于对照组(P〈0.05),实验组神经轴突总数,有髓轴突总数高于对照组(P〈0.01),实验组Ⅰ型胶原(ColⅠ)沉积和Ⅲ型胶原(ColⅢ)的沉积比对照组少(P〈0.05)。结论在体内环境下维拉帕米能抑制成纤维细胞的生长及分泌细胞外基质,抑制ColⅠ和ColⅢ的分泌,从而减少瘢痕组织的形成,促进神经轴突的生长。Objective To observe the preventive effect of verapamil on scar formation after peripheral nerve injury. Methods The end-to-end anastomosis model of the transected sciatic nerve was established in 60 healthy male SD rats. The rats were randomly divided into experimental group ( right side) and control group ( left side). The experimental group was wrapped with a piece of gelfoam saturated with vera- pamil, and the control group with a piece of gelfoam soaked with normal saline. Rats were killed 6 weeks later, and the transected parts of the nervous tissue were examined by using histology, Western blotting and immunohistochemistry. The muscular wet weight was measured, and the muscle atrophy index was analyzed. Results The experimental group exhibited heavier muscularis wet weight and lower muscle atrophy index than the control group ( P 〈 0. 05 ) , and the number of axons, including the total number of myelinated axons, in the experimental group was significantly greater than in the control group (P 〈 0. 01 ). The collagen Ⅰ (Col Ⅰ) and collagen Ⅲ(Col Ⅲ) deposition in the experimental group was less than in the control group (P 〈 0.05). Conclusion In vivo verapamil can inhibit fibroblasts to secrete Col Ⅰ and CoⅠ Ⅲ, decrease scar formation and promote the growth of neurites.
