桉油精对补骨脂素体外抗肿瘤活性的增效作用研究

摘　　要：目的考察桉油精对补骨脂素体外抗肿瘤活性的增效作用。方法采用MTT法观察桉油精与补骨脂素对4种人胃癌细胞的生长抑制作用,计算IC50,选择对补骨脂素最为敏感的一种癌细胞,以确定桉油精与补骨脂素的最佳比例与浓度。结果补骨脂素作用于SGC-7901、MKN-45、BGC-823、MGC-803的IC50分别为36.23、66.26、36.9、40.27μg/mL,白蔻仁挥发油及桉油精单用无显著抗肿瘤作用,但其与补骨脂素联用,有显著的抑制增强作用。选用对补骨脂素最为敏感的SGC-7901进一步研究。结论桉油精对补骨脂素的体外抗肿瘤活性具有协同作用。桉油精与补骨脂素配伍的最佳抑制比例与剂量为2∶1,72∶36μg/mL。AIM To investigate the synergistic effect in vitro of eucalyptol combined with psoralen on the anti- tumor activity. METHODS MTT method was used to observe the cell growth inhibition of eucalyptol and psor- alen monomer on four kinds of human gastric carcinoma cell lines and calculate the IC50 values. The optimal propor- tion and concentration were determined by choosing a kind of cancer cells most sensitive to psoralen. RESULTS The IC50 of psoralen to human gastric carcinoma cell line MGC-803, BGC-823, SGC-7901 and MKN-45 was 40. 27, 36.9, 36.23, 66.26 μg/mL, respectively. Single use of volatile oil from Ammomi Fructus rotundus and eucalyptol had no effect on anti cancer. But the combination with eucalyptol and psoralen presented significant inhi- bition enhancement. Cell line SGC-7901, most sensitive to psoralen, was chosen to research the optimal proportion of eucalyptol and psoralen. CONCLUSION Eucalyptol and psoralen in vitro have synergistic effect on the antitu- mor activity with best inhibition ratio of 2 ： 1 and concentration of 72 ： 36 μg/mL.
