摘  要:目的:建立高效液相色谱法测定肋柱花中獐牙菜苦苷和当药黄素含量的方法。方法:采用高效液相色谱法测定肋柱花中獐牙菜苦苷和当药黄素的含量。色谱柱为Phenomenex C18(4.6 mm×250 mm,5μm),流动相为水(A)和甲醇(B)梯度洗脱,流速1.5 mL.min-1,柱温25℃,检测波长237 nm。结果:獐牙菜苦苷对照品在0.27~5.40 mg与峰面积呈现良好的线性关系(r=0.999 8),加样回收率为98.49%、RSD 1.06%(n=9);当药黄素对照品在0.165~3.3 mg与峰面积呈现良好的线性关系(r=0.999 6),加样回收率为97.35%、RSD 1.45%(n=9)。结论:该方法简单、快捷、准确、重复性好,可用于肋柱花中獐牙菜苦苷和当药黄素的含量测定。Objective: To establish an HPLC method for the simultaneous determination of swertiamain and swertisin in herba Lomalogonium rotatum. Method: Swertiamain and swertisin were simultaneously analyzed using HPLC accompanied with Phenomenex C18 (4.6 mm×250 mm, 5 μm) column and UV detector. Methanol-water was chosen as the mobile phase with gradient elution. The wavelength was set at 237 nm, while the flow rate was set at 1.5 mLmin-1. Result: Swertiamain and swertisin were linear within the range of 0.27-5.40 mg (r=0.999 8), 0.165-3.3 mg (r=0.999 6), respectively. Meanwhile, the average recoveries were 98.49% (RSD 1.06%) and 97.35% (RSD 1.45%), respectively. Conclusion: The method is simple, rapid, accurate and possesses good reproducibility. It can be used for the quality control of Lomalogonium rotatum.
