摘  要:目的:以腹主动脉缩窄造成压力负荷超载的大鼠心力衰竭模型为研究对象,观察压力负荷超载性心衰发生发展过程中心肌能量代谢及心脏功能的变化及坎离颗粒的影响。方法:采用腹主动脉缩窄法制成心脏压力负荷超载心衰模型,造模1周后灌胃给药,以培哚普利(5.4 mg.kg-1)和曲美他嗪(0.36 mg.kg-1)作为阳性对照药,观察坎离颗粒各组(13.5,6.75,3.38 mg.kg-1)给药32周后,血流动力学指标及心肌能量代谢的变化。结果:与假手术组比较,模型组大鼠左室收缩压(LVSP,P<0.01)和左室舒张末期压(LVEDP,P<0.05)均显著升高,+dp/dtmax(P<0.01)和-dp/dtmax(P<0.05)均加大,三磷酸腺苷(ATP),ATP/二磷酸腺苷(ADP)显著降低(P<0.001);与模型组比较,坎离颗粒3个剂量组LVEDP均显著降低(P<0.05),坎离颗粒中剂量组+dp/dtmax则有加大,且明显大于培哚普利组(P<0.01),坎离中剂量组ATP显著增高(P<0.05)。结论:坎离颗粒干预显著改善该心衰模型心肌能量代谢并同时改善左室舒张功能。Objective: To study the influence of Kanli granule on myocardial energy metabolism and cardiac function. Method: Heart failure model were developed in rats by transverse aortic constriction(TAC). After 1 week of the operation, rats were treated orally with trimetazidine(0.36 mgkg-1),perindopril(5.4 mgkg-1),Kanli granule high-medium-low dose(13.5,6.75,3.38 mgkg-1). After 32 weeks, myocardial energy metabolism and hemodynamic were examined. Result: Compared with group sham,LVSP(P〈0.01) and LVEDP(P〈0.05) was increased in model group,+dp/dtmax(P〈0.01) and -dp/dtmax(P〈0.05) was large in group BD, ATP,ATP/ADP was reduced (P〈0.001) in model group BD;Compared with model group,LVEDP(P〈0.05) were all reduced in group of Kainli granule,+dp/dtmax(P〈0.05),ATP was increased(P〈0.05) in group M of Kainli granule. Conclusion: Kanli granule can improve left ventricular diastolic function, energy metabolism and improve the efficacy of cardiac structure and function.
