摘  要:目的对心肌梗死后心力衰竭大鼠经尾静脉注射腺病毒介导的β肾上腺素受体激酶抑制剂(βARKct),观察其对血流动力学的影响。方法体重250 g左右的雄性SD大鼠随机分为3组,其中两组结扎冠状动脉左前降支形成急性前壁心肌梗死,4周后形成心力衰竭:对照组心衰大鼠尾静脉注射空腺病毒载体,实验组心衰大鼠尾静脉注射腺病毒介导的肾上腺素受体激酶抑制剂;另一组为假手术组尾静脉注射空腺病毒载体。4周后测定各组大鼠左室射血分数及血流动力学指标。结果与假手术组相比,对照组及实验组大鼠左室射血分数下降(P<0.05),左心室内压力变化最大速率(±dp/dt max)绝对值下降(P<0.05);与对照组相比,实验组大鼠左室射血分数升高(P<0.05),左心室内压力下降最大速率(-dp/dt max)绝对值升高(P<0.05)。结论通过尾静脉注射腺病毒介导的肾上腺素受体激酶抑制剂(Ad-βARKct)可以明显改善心肌梗死后心力衰竭大鼠的射血分数及血流动力学指标。Objective To study the effect of adenovirus - mediated inhibitor of [5 - adrenergic receptor kinase through caudal vein on haemodynamics of rats with heart failure after myocardial infarction. Methods The male SD rats with body weight of 250 grams were divided into three groups. Rats in 2 groups underwent ligation of left anterior descending (LAD) branch of coronary artery to induce acute myocardial infarction, and heart failure occurred after 4 weeks, and rats with heart failure in control group were injected with empty adenovirus vector through the caudal vein, and rats with pseudo - operation in experimental group were injected with Ad -βARKct through the caudal vein; and rats with heart failure were injected with empty adenovirus vector through the caudal vein. Left ventricular ejection fraction (LVEF) and index of hemodynamics were measured 4 weeks later. Results Compared with rats in sham - operated group, LVEF was lower ( P 〈 0.05 ), and + dp/dt max was also lower in rats of both control group and experimental group ( P 〈 0. 05 ). Compared with rats in control group, LVEF was higher ( P 〈 0.05 ), and - dp/dt max was also higher in rats in experimental group ( P 〈 0.05 ). Conclusion In vivo, delivery of Ad - βARKct can improve cardiac function and index of hemodynamic in rats with heart failure after myocardial infarction.
