磷脂酰乙醇胺结合蛋白过表达CHO细胞的建立及对细胞周期的影响

摘　　要：目的建立过表达磷脂酰乙醇胺结合蛋白(PEBP)的CHO细胞,并研究PEBP过表达对CHO细胞周期的影响。方法将PEBP克隆于潮霉素B抗性的表达质粒,转染CHO细胞,潮霉素B压力下筛选细胞单克隆,流式细胞法检测细胞周期。结果建立了与内源性表达量相当的,可稳定过表达外源性PEBP的CHO细胞,与对照细胞克隆比较,过表达PEBP的CHO细胞周期无显著变化。结论 PEBP过表达没有导致CHO细胞周期的改变。Objective To establish a CHO cell line with overexpressing phosphatidylethanolamine - binding protein （PEBP）, whose possible effect on cell cycle was also been investigated. Methods Cloning PEBP into a expression plasmid with hygromycin B resistance, following by stable transfection to CHO cells, and then monoclone was screened with the existence of hygromyein B. Cell cycle was determined by flow eytometry. Results A cell line expressing exogenous PEBP equal to endogenous PEBP was established, and the cell cycle of PEBP over - expressing cells were not change compared to control clone. Conclusion PEBP over - expression did not effect cell cycle of CHO cells.
