摘 要:目的研究抗栓中药血栓心脉宁片(XXT)对人脐静脉内皮细胞(HUVEC)基因表达谱的影响,探讨XXT对血管内皮细胞作用的分子机制。方法 XXT与HUVEC共培养24h,运用Bio 35khuman Genome Array全基因组表达芯片,检测XXT对HUVEC基因表达谱的影响,分子注释系统MAS3.0软件进行聚类分析。荧光定量PCR(Real Time–PCR)验证基因表达谱结果。结果 XXT作用内皮细胞24h后,基因芯片分析筛选出差异表达大于1.5倍的基因2 130个,其中上调基因689个,下调基因1 399个,包括转录调节、炎症反应、免疫反应、细胞黏附、抗凋亡、信号转导等相关基因,经Real Time-PCR验证结果与基因芯片结果一致。结论 XXT可影响血管内皮细胞多条细胞信号传导通路相关的基因表达,如参与细胞凋亡、氧化应激和抗凝等多个基因。Objective To investigate the effects of Xueshuan Xinmaining tablet(XXT) on gene expression profiles of human umbilical vein endothelial cells(HUVECs) ,and to explore the molecular mechanisms of XXT on vascular endothelial cells. Methods Af ter XXT acted on HUVECs for 24 h,Bio 35k human genome array oligonucleotide microarray was applied to detect the alteration of gene expression profiles induced by XXT in HUVECs. The cluster analysis was performed by molecular annotation system MAS 3.0 software. Real time PCR (RT - PCR) was used to verify the resuk of selected differentially expressing genes. Results The total 2130 genes,including 689 upregulated and 1399 clownregulated genes, were obtained with differential expression more than 1.5 -fold after XXT acted on HUVECs for 24 h. XXT affected the expression levels of genes involved regulation of gene transcription,inflam- mation,immune response,cell adhesion,anti- apoptosis, signal transduction and so on. The results verified by RT- PCR consistent with the microarray results. Conclusion XXT modulate function of endothelial cells by regulating sets of genes through different cellular signal transduction pathway involved in cell apoptosis,oxidative stress,and anticoagulation.
