摘 要:目的探讨不同价态砷亚慢性暴露对大鼠肝脏抗氧化作用及脂质过氧化水平的影响。方法 Wistar大鼠70只,按体重随机分成正常对照组、亚砷酸钠(iAs3+)低剂量组、中剂量组和高剂量组,砷酸氢钠(iAs5+)低剂量组、中剂量组和高剂量组,每组10只,不同价态砷高、中、低染毒剂量均为20.00mg/kg、6.67mg/kg、2.33mg/kg,对照组自由饮用不含砷蒸馏水。连续染毒90d,采用相关试剂盒方法检测大鼠肝脏组织中脂质过氧化产物丙二醛(MDA)、GSH和GSSG含量,并计算GSH/GSSG比值以及谷胱甘肽S-转移酶(GST)和谷胱甘肽过氧化物酶(GSH-Px)活力。结果随不同价态砷染毒剂量的增加,大鼠肝脏组织中MDA含量增加(P﹤0.05),GSH-Px活力降低(P﹤0.05);iAs5+高、中剂量组GSH-Px活力均相应高于iAs3+同一剂量组,iAs5+高、中剂量组MDA含量均相应低于iAs3+同一剂量组。iAs3+和iAs5+各剂量组GST活性和GSH含量剂量-效应关系较为明显,均随剂量增加而下降;iAs5+高、中剂量组GST活性及GSH含量均高于iAs3+组(P﹤0.05);iAs5+高、低剂量组GSSG浓度均高于iAs3+组(P﹤0.05);不同价态砷各剂量组GSH/GSSG比值均随剂量增加而降低,iAs3+组的下降趋势更为明显。结论不同价态砷亚慢性暴露均可使大鼠肝脏脂质过氧化水平增强,抗氧化能力降低;iAs3+对肝脏抗氧化能力的抑制作用强于iAs5+,iAs3+对肝脏的脂质过氧化损伤作用要较iAs5+严重。OBJECTIVE To know the effects of subchronic exposure to arsenite and arsenate on antioxidation and lipid peroxi- dation in liver of rat. METHODS Seventy Wistar rats were randomly divided into seven groups including control group (C), iAs^3+ low, middle, high dose groups, iAs^5+ low, middle, high dose groups, 10 in each. Treated with sodium arsenite and sodium arsenate through drinking water at the dose of 0, 2.33, 6.67 and 20.0 mg/kg respectively for 90 consecutive days. After 90 days of the treatment, the contents of malondialdehyde (MDA), GSH and GSSG were measured by ELISA kit, and the ra- tio of GSH/GSSG were measured. The activities of glutathione peroxidase (GSH-Px) and glutathione S-transferase (GST) in the liver tissues were determined by ELISA kit method. RESULTS The results showed that with the increase of exposure to ar- senite and arsenate, the content of MDA significantly increased and the activities of GSH-Px significantly decreased with a dose-dependent manner (P 〈 0.05) ; The activities of GSH-Px in iAs^5+ high and middle dose groups were higher than the same iAs^3+ groups (P〈 0.05), the content of MDA in iAs^5+ high and middle dose groups were lower than the same iAs^3+ groups (P〈 0.05 ). With the increase of exposure to arsenite and arsenate, the activity of GST and the content of GSH significantly decreased with a dose-dependent manner (P 〈 0.05). The activities of GSH-Px and the content of GSH in iAs^5+ high and middle dose groups were higher than the same iAs^3+ groups (P〈 0.05) ; The content of GSSG in iAs^5+ high and low dose groups was higher than the same iAs^3+ groups (P 〈 0.05). With the increase of exposure to arsenite and arsenate, the ratio of GSH/GSSG signifi- cantly decreased with a dose-dependent manner (P 〈 0.05 ), it showed a significant descend trend in iAs^3+ groups.
