摘 要:目的本研究探讨体外培养大鼠胚胎皮肤成纤维细胞和人胚胎皮肤成纤维细胞内Ⅰ、Ⅲ型胶原的表达及意义。方法分别从大鼠胚胎(孕14d)和人胚胎(孕20周),获得原代的胚胎皮肤成纤维细胞。将第三代细胞荧光量子点标记技术检测其内Ⅰ、Ⅲ型胶原的表达。结果培养的第三代大鼠胚胎皮肤成纤维细胞和人胚胎皮肤成纤维细胞具有相同的显微结构,量子点标记检测其I、Ⅲ型胶原表达的平均光密度比较,差异无统计学意义(P﹥0.05)。结论体外培养第三代大鼠胚胎皮肤成纤维细胞和人胚胎皮肤成纤维细胞具有相同的形态结构及合成I、Ⅲ型胶原的能力相似,因此可能在组织无瘢痕愈合的修复过程中发挥相同的功能。OBJECTIVE To explore the significance of the expression of type collagen Ⅰ、Ⅲ in the cultured fibroblasts of rat embryonic skin and human festal skin fibroblasts. METHODS The primary fibroblasts were isolated from the rat embryos (ges- tation 14 days) and human embryos (gestation 20 weeks). The expression of type Ⅰ、Ⅲ collagen in the third generation of cells were detected by fluorescent quantum dots. RESULTS The third generation of MEFB (mouse embryonic skin fibroblast) and HEFB (human embryonic skin fibroblasts) possessed the same microstructure; There was no significant difference in the aver- age optical density between the expression of type I and III collagen marked by the quantum dots (P 〉 0.05). CONCLUSION The third generation of EEFB and HEFB has the same morphology ; The synthesis ability of type Ⅰ、Ⅲ collagen is similar. It is suggested that EEFB and HEFB may play the same role in the tissue repair process of scarless healing.
