保康柴胡种质资源的ISSR研究

摘　　要：目的通过ISSR分子标记技术考察保康柴胡与不同品种柴胡间的亲缘关系。方法从60条引物中筛选出7条合适的引物,对不同品种的柴胡进行ISSR分析,构建聚类系统树状图。结果 7条引物共扩增出81条条带,其中多态性条带79条,占97.5%,聚类分析显示同产地内遗传特性相对稳定,但种内遗传变异明显。讨论保康安家湾柴胡种质较好,值得推广。Objective To assess population genetic diversity of different kinds of individuals of Bupleurum by inter simple sequence repeat （ISSR） technique. Methods Seven appropriate ones were selected out from a total of sixty primers for ISSR PCR amplification and draw dendrograms by using genetic distance UPGMA method. Result A total of 81 bands and 79 polymorphic bands were amplified by 7 primers, The cluster analysis showed that genetic diversity was obvious in Bupleurum L. Conclusion The germplasm of Thorowax collected from Baokang was good and was worth popularizing.
