摘 要:目的研究姜黄素对急性脊髓损伤的保护作用及其可能机制。方法采用改良Allen’s打击法建立大鼠急性脊髓损伤模型,将60只SD雌性大鼠随机平均分为假手术组、模型组、姜黄素低剂量组(40mg/kg,CUR-L组)、姜黄素高剂量组(100mg/kg,CUR-H组),每组各15只。术后7d采用脊髓损伤后后肢运动功能评分系统(BBB法)评测大鼠后肢运动功能;化学比色法检测大鼠脊髓组织中丙二醛(MDA)含量变化;蛋白质印迹(Western blot)及免疫组化检测核因子E2相关因子2(Nrf2)、γ-谷氨酰半胱氨酸合成酶(γ-GCS)蛋白表达;苏木精-伊红(HE)染色观察脊髓组织的病理改变。结果与模型组相比,姜黄素组大鼠后肢功能明显改善;脊髓组织中MDA含量降低;Nrf2、γ-GCS蛋白的表达上调;脊髓组织病理损伤减轻。结论姜黄素对大鼠急性脊髓损伤有保护作用,其机制可能是通过激活Nrf2/抗氧化反应原件(ARE)通路从而上调γ-GCS蛋白表达来实现的。Objective To investigate the protective effects of curcumine on acute spinal cord injury and its possible mechanism. Methods Rat models of acute spinal cord injury were established by modified Mlen' s as- say. Totally 60 female rats were randomly divided into four groups: sham-operation group, model group, low-dose curcumine group { 40mg/kg, CUR-L group } and high-dose eurcumine group { 100mg/kg, CUR-H group }. At 7 days after operation,motor function of the rat hind limb was determined by the BBB{ Basso Beattie and Bresnaban) ,con- tent of Malondialdehyde { MDA } in spinal cord tissues was detected with chemical eolorimetry, and Western blot and immunohistoehemistry analysis were employed to detect the Nrf2, ~,-GCS protein expression;pathological changes of brain tissues were observed with HE staining. Results Compared with the model group,hind limb function was significantly improved;MDA content was decreased in the spinal eord;Nrf2 and Y-GCS protein expression was in- creased; and the pathological injury of spinal cord was relieved in the eurcumine groups. Conclusion Cureumine has the protective effects on acute spinal cord injury. The mechanism may be regulated by activating Nrf2/ARE pathway to up-regnlate Y-GCS protein expression.
