摘 要:目的观察人骺板软骨细胞与丝素支架在体外进行共培养的生物相容性。方法将第一代人骺板软骨细胞与可降解的多孔丝素材料(支架)在体外进行复合、培养。用倒置显微镜、透射电子显微镜、激光共聚焦显微镜和MTT等方法,观察细胞与丝素材料在体外的生物相容性。结果人骺板软骨细胞在可降解的多孔丝素材料上生长良好,随培养时间的增加而大量增多。结论人骺板软骨细胞与可降解的多孔丝素材料在体外具有良好的生物相容性,有希望作为构建组织工程骺板理想的细胞和支架。Objective To observe the biocompatibility of human epiphyseal plate chondrocyte on porous silk scaffold in in-vitro culture. Methods The morphology of first passage of human epiphyseal plate chondrocytes grown on porous silk scaffold was observed by inverted microscope, transmission e- lectron microscope and laser scanning confocal microscope, and the cell viability was determined using the MTT test. Results The first passage of human epiphyseal plate chondrocytes observed under the inverted microscope were fusiform in shape after 2 days and grew to confluence in one well of a six- well plate in 3 days. Plenty of the rough endoplasmic reticula were observed within the chondrocytes under transmission electron microscope. On the other hand, chondrocytes grown on scaffold were visi- ble within 2 days in culture and grew in abundance after 8 days when observed under the laser scanning confocal microscope. There was no significant difference in cell viability between the first passage of human epiphyseal plate chondrocytes in in-vitro culture group and porous silk scaffold group using MTT test (Paired Samples Test, t = 1. 871, P = 0. 094, P 〉 0. 05). Conclusions Human epiphyseal plate chondrocytes have good biocompatibility in in-vitro culture using porous silk scaffolds, and may be used in epiphyseal plate tissue engineering.
