摘 要:[目的]通过体外实验评价本课题组自主研发的新型低模量β钛合金的生物相容性。[方法](1)采用非自耗真空电弧炉熔炼Ti35Nb3Zr2Ta,应用扫描电子显微镜(SEM)、原子力显微镜(AFM)、能谱扫描(EDX)和表面接触角测量方法观察材料表面特征;(2)将原代培养的大鼠成骨细胞与实验组(Ti35Nb3Zr2Ta)和对照组(Ti6Al4V)合金体外培养,通过细胞计数、细胞骨架染色、MTT比色法、碱性磷酸酶活性测定、逆转录聚合酶链反应(RT-PCR)和茜素红染色等方法观察成骨细胞在两组钛合金表面的粘附、伸展、增殖、分化及成骨能力。[结果]材料表面特征测试结果显示Ti35Nb3Zr2Ta具有比Ti6Al4V更高的表面粗糙度和更好的亲水性。细胞实验表明成骨细胞在Ti35Nb2Ta3Zr表面的粘附、伸展、增殖、分化优于Ti6Al4V。[结论]新型低模量β钛合金Ti35Nb3Zr2Ta显示了良好的生物相容性,有望成为一种新的人工关节假体。[ Objectivel To evaluate the in vitro biocompatibility of a novel [3 type titanium alloy with a low Young's modulus. [ Method] (1) The Ti35Nb2Ta3Zr alloy fabricated in a vacuum consumable arc melting furnace were tested with SEM, EDX,AFM,contact angel meter. (2) Osteoblasts were cultured on experimental group (Ti35Nb3Zr2Ta) and control group (Ti6A14V) in vitro. The adhesion, spread, proliferation, differentiation and mineralization of osteoblasts were observed by cells counting, cytoskeleton dyeing, standard MTT assay, reverse transcriptase - polymerase chain reaction ( RT - PCR) analysis and alizarin red staining method. [ Result] The Hydrophilicity and Ra value of Ti35Nb2Ta3Zr was significantly higher than that of Ti6AI4V. The in vitro experiments showed that osteoblasts attached, spreaded, proliferated, and differentiated better on Ti35Nb2Ta3Zr than on Ti6A14V. [ Conclusion] Since the novel β type titanium alloys(Ti35Nb3Zr2Ta) seem to possess good biocompatibility in vitro, it may be a new type of material as joint prosthesis.
