摘 要:对分离的 2 0株鸡致病性大肠杆菌进行了药敏实验 ,筛选出 1 3株耐氧氟沙星 ( OFL )的菌株。经质粒提取、电泳和转化实验 ,获得一株由质粒介导的耐 OFL的多重耐药菌株。该菌株对参试的 1 1种抗生素全部耐药 ,其中 OFL MIC>50 μg/ml。该菌 R质粒电泳可见 6条带 ,转化大肠杆菌 DH5α,在 2 0 μg/ml OFL的 LB平板筛选得到转化子。该转化子含有原菌株中 2 .7kb的质粒 ,获得原菌株部分耐药性。连续转化 5次 ,均获得耐 OFL的转化子。证实该菌株对Thirteen ofloxacin resistant strains were collected from twenty clinical isolates of chicken pathogenic E.coli through the drug susceptibility test. After the plasmid DNA extraction and agarose gel electrophoresis test, the resistant plasmids was introduced into E.coli DH5α by transformation. Ofloxacin resistant bacterial colonies were selected on the agar LB plates containing OFL (20μg/ml). As a result, only one multi resistant strain whose ofloxacin resistance is associated with its plasmid DNA was obtained. This strain with a MIC of OFL of >50 μg/ml is resistant to the eleven antibiotics used in the test. The resistance to ofloxacin could be stably reserved in generation after generation through the ofloxacin resistant plasmid whose molecular weight was about 2.7kb.
