摘 要:目的 ICP-MS法研究纳米银在小鼠肺内和肺外脏器的含量。方法制备Sigma纳米银颗粒,气管滴注法纳米银染毒小鼠。染毒后,在不同的时间点,处死小鼠,留取肺脏、肝脏、肾脏、脾脏和心脏,电感耦合等离子体-质谱法(ICP-MS)测量各个器官内纳米银的含量。结果气管滴注进入小鼠体内的纳米银,很快随血流分布到全身各脏器,滞留在体内的纳米银主要集中在肺、肝、脾、肾等器官,体内纳米银浓度随着滴注浓度成正相关。低、中、高浓度的纳米银银染毒第1天测量肺脏纳米银的含量分别是(1.64±0.271)ng/mg,(8.581±1.503)ng/mg和(16.196±1.503)ng/mg,明显高于对照组(P<0.01)。高浓度纳米银染毒后,纳米银肝脏中含量从第1天的(0.272±1.670)ng/mg增高到第28天的(1.124±0.248)ng/mg,均明显高于对照组(P<0.01);脾脏纳米银浓度从第1天的(0.470±0.060)ng/mg降低到第28天的(0.265±0.056)ng/mg,明显高于对照组(P<0.01);肾脏纳米银的含量第1天最高为(0.201±0.008)ng/mg,明显高于对照组(P<0.01);心脏染毒第1天纳米银的含量(0.155±0.005)ng/mg,明显高于对照组(P<0.01)。结论气管滴注纳米银颗粒,肺内和肺外重要脏器均能检测到纳米银。Objective Study the content of nano-silver in the lung and extrapuhnonary organs of ,nice with the ICP-MS method. Methods Sigma nano-silver was used by different concentrations with int,'atracheal instillation into the mice,and the important viscera were taken out at different time,and the 1CP-MS method was used to measure the content of nano-silver in these organs. Results Instilled nano- silver particles were transloeated into the organs via hloodstream immediately,and deposited mostly in lung,liver,spleen and kidney. With the increasing concentration of nano-silver instilled into the mice,the content uf nano-silver in the organs increased. The content of nano-silver of hmg in low, medium and high concentration group at 1 th day was ( 1. 647 ± 0.271 ) ng/mg, ( 8.581 ± 1. 503 ) ng/mg and ( 16. 196 ± 1. 503 ) ng/mg respectively, which was higher than in the control group signifieantly(P 〈 0.01 ) ;The eontent of nano-silver of liver in high concentra- tion group from (0. 272 ± 1. 670 ) ng/mg at 1 d increased tn ( 1. 124 ± 0. 248 ) ng/mg at 28d, hut was still higher than that in the control group significantly ( P 〈 0.01 ) ; The content of nann-silver of spleen in high concentration group from ( 0. 470 ± 0.060 ) ng/mg at I d decreased to ( 0. 265 ± 0.056 ) ng/mg at 28d, hut was still higher than that in the control group significantly ( P 〈 0.01 ) ;The highest content of nano-silve," of kidney in high concentration gronp at I d was ( 0.200 ± 0. 008 ) ng/mg, higher than that in the control group significantly ( P 〈 0.01 ) ; The con- tent of nano-silver of heart in high concentration group at l d was(0. 155 ± 0. 005)ng/mg, higher than that in the control group significantly( P 〈0.01 ). Conclusion The experiment indicates that the nano-silver particles can be measured in the lungs and extrapulmonary organs.
