摘 要:为了探究外源NO对半夏体内次生代谢的调控机制,以半夏组培苗为材料,研究在喷洒不同浓度SNP(NO供体)后体内苯丙氨酸解氨酶(PAL)活性、谷氨酸合酶(GOGAT)活性、谷草转氨酶(GOT)活性、总生物碱质量分数和鸟苷质量分数的变化和规律.结果表明:不同浓度SNP均能提高半夏组培苗体内苯丙氨酸解氨酶(PAL)、谷氨酸合酶(GOGAT)、谷草转氨酶(GOT)活性,增加次生代谢产物总生物碱和鸟苷质量分数.其中,1mmol/L SNP处理组在第24天时效果最好,总生物碱及鸟苷质量分数分别比对照增加了66.1%,188.3%.外源NO可能通过提高半夏组培苗体内PAL,GOGAT,GOT活性,加速无机氮的同化及转化效率,提供更多的代谢前体物以促进总生物碱及鸟苷积累.In this paper, the effects of exogenous NO on the activities of several secondary metabolism key enzymes and the contents of total alkaloids and guanosine in regenerated Pinellia ternate plantlets werestudied. The results showed that exogenous NO enhanced the activities of PAL (phenylanlanine ammonia- lyase), GOT (glutamic-oxaloacetic transaminase) and GOGAT (glutamate synthase) and increased thecontents of total alkaloids and guanosine of regenerated P. ternate in different concentration treatments, of which 1 mmol/L SNP (the donor of NO) gave the best results and increased alkaloids and guanosine con-tents by 66.2% and 190.9%, respectively. Exogenous NO promoted the activities of PAL, GOGAT and GOT in regenerated P. ternate plantlets, which accelerated the assimilation and conversion efficiency ofinorganic nitrogen and furnish more precursor of secondary metabolism. Furthermore, the contents of alkaloids and guanosine increased correspondingly.
