摘 要:核质蛋白是广泛存在于卵母细胞中的由Npm基因编码的一种酸性、磷酸化的蛋白质,为分子伴侣家族的一员。本研究通过RT-PCR检测成熟不同时期卵母细胞Npm mRNA基因表达,同时采用免疫组化以及Western blot检测成熟不同时期卵母细胞核质蛋白定性和定量表达。结果:成熟不同时期卵母细胞中均可检测到Npm基因mRNA表达。采用免疫组化的方法在体外成熟0 h和11 h卵母细胞中可检测到核质蛋白表达,但成熟22 h、33 h、44 h的卵母细胞检测不到核质蛋白表达;采用Western blot技术在体外成熟0 h和11 h卵母细胞中检测不到核质蛋白,成熟22 h、33 h、44 h卵母细胞可检测到核质蛋白。结果说明随着卵母细胞体外成熟进程的推进,Npm基因及其编码的核质蛋白表达量不断增加,可为受精后精子核解聚或体细胞核重编程做准备。Nucleoplasmin, the most abundant protein in the oocyte, is an acid and phosphorylated protein and belongs to the archetypal nu- clear chaperone. In this paper, RT-PCR was used to detect the expression of Npm gene of different in vitro maturation periods of oocytes. The results showed that the expression of Npm gene could be detected in different periods oocytes. Immunocytochemistry was used to detect the expression of nucleoplasmin of different in vitro maturation periods of oocytes. The results showed that nucleoplasmin expression could be de- tected in Oh and 1 lh groups, but not detected in 22h, 33h and 44h groups. However, nucleoplasmin expression could not be detected in Oh and 1 l h groups, but detected in 22h, 33h and 44h groups by western blotting. These results indicated that as the oocyte maturation process forward, the expression of Npm gene and its coded nucleoplasmin were increased and could facilitate the decondensation of sperm chromatin or reprogramming of somatic cell nucleus.
