摘 要:[目的]通过短期小剂量吲哚美辛灌胃制备大鼠非甾体类抗炎药(Non-Steroid Anti-Inflammatory Drugs,NSAIDs)相关急性小肠损伤模型,研究云母颗粒对大鼠NSAIDs相关急性小肠损伤的保护作用并探讨其可能作用机制。[方法]将50只SD雄性大鼠按随机数字表法平均分成正常对照组、模型组、云母组、米索前列醇组、蒙脱石组5组。对照组予生理盐水2mL/qd灌胃;模型组予生理盐水2mL、云母组予云母颗粒混悬液0.9g.kg-1、米索前列醇组予米索前列醇混悬液60μg.kg-1、蒙脱石组予蒙脱石混悬液0.9g.kg-1灌胃,给药0.5h后各组予吲哚美辛溶液7.5mg.kg-1qd灌胃,共给药5天后处死。[结果]模型组大体及组织学损伤评分、血浆D-乳酸含量、肠系膜淋巴结(mesenteric lymph node,MLN)细菌移位(bacterialtranslocation,BT)阳性率及含菌量显著升高(与正常对照组比,P<0.05);云母组与模型组比,各指标显著降低(P<0.05);米索前列醇组和蒙脱石组BT阳性率与模型组比无显著性差异(P>0.05),但两者MLN的含菌量均较模型组明显下降(P<0.05);MLN含菌量与血浆D-乳酸含量、大体及组织学损伤评分呈正相关(P均<0.01)。[结论]吲哚美辛灌胃能够诱导大鼠NSAIDs相关急性小肠损伤,这可能与黏膜通透性升高致BT,进一步加重小肠黏膜损伤有关;云母颗粒可能通过降低黏膜通透性、拮抗BT而对大鼠NSAIDs相关急性小肠损伤起到保护作用。[Objective] To establish the rat model of NSAIDs-induced small intestinal acute injury by oral administration of indomethacin and to observe the protective effect of YunMu particle on the small intestinal injury, and to explore the possible mechanism of the protective effect of YunMu particle. [Methods]50 healthy male SD rats were randomly and equally divided into five groups:(1)Normal control group; (2)Model group; (3)YunMu group;(4) Misoprostol group; (5)Smectite group. Normal control group received distilled water lmL/(100 god^-1) intragastrically for 5 days; Model group received indomethacin 7.5 mg.kgod^-1 intragastrically for 5 days; and three treatment groups receivied YunMu particle suspension 0.9g.kgod^-1, misoprostol suspension 60ug.kgod^-1, smectite suspension 0.9g.kg.d ^-1 intragastrically for 5 days, respectively, beginning from the day before indomethacin was given as Model group. [Results]Morphological and histological injury score, plasma concentration of D-Lactate, bacterial translocation(BT) rate of MLN were higher than Normal control group; all kinds of indexes of YunMu group were significantly lower than Model group; BT rate of Misoprostol group and Smectite group were not significantly lower than Model group,but numbers of enterobacteria of MLN of Misoprostol group and Smectite group were significantly lower than Model group; there was positive correlation between plasma concentration of D-Lactate,morphological injury score,histological injury score and numbers of enterobacteria of MLN. [Conclusion] Indomethacin administered intragastrically can establish rat model of NSAIDs-induced small intestinal a- cute injury and the mechanism is possibly related to increased mucosal permeability, bacterial translocation; NSAIDs-induced small intestinal acute injury of rats can result in BT of MLN which has positive correlation with mucosal permeability and BT can aggravate the small intestinal injury; YunMu particle has protective effects on NSAIDs-induced small intestinal a
