摘 要:为获得高效纤维素降解菌株,以CMC-Na为唯一碳源进行筛选,从土壤及腐烂秸秆中分离到26株纤维素降解菌。分别测定其滤纸崩解、CMC-刚果红水解圈、CMCase(羧甲基纤维素酶)和FPase(滤纸片酶)活性等指标,从中筛选出产纤维素酶能力最强的JSD-1放线菌。通过测定不同条件下JSD-1放线菌产纤维素酶的活性,得到其最佳产酶条件。结果表明,当温度为35℃、发酵液初始pH为6.5、接种量为8%及转速为180r/min时,CMCase和FPase分别在发酵第5天和第4天有最大的产酶活性,为59.19和31.68U/mL。In order to obtain microbes with high quality of cellulose-decomposing enzyme, twenty six strains of cellulose-decomposing microbes were isolated from soil and rotten straw by using the media supplemented with CMC-Na as the sole carbon resource. For these strains,the following experiments were conducted incIuding the decomposition of filter-paper, decoloration of Congo red, production of CMCase and FPase. The actinomycete JSD-1 was selected due to its strongest ability to degrade cellulose and conditions for cellulase production of JSD-1 are also established. The results showed that the optimum cultural temperature for JSD-1 was 35℃ ,the pH for initial fermentation is 6. 5, the inoculation volume is 8%, the rotate speed is 180 r/min and the suitable culture time is 5 and 4 d for the production of CMCase and FPase, respectively. Under these conditions,the activity of CMCase and FPase reached at 59.19 and 31.68 U/mL, respectively.
