摘 要:目的探讨食管鳞癌组织中RhoC、Bmi-1基因表达及其对预后的影响。方法采用免疫组织化学方法检测RhoC、Bmi-1基因在80例食管鳞癌患者中的表达,应用检验、COX回归模型和Kaplan-Meier生存曲线检测分析其与患者预后的关系。结果在食管癌组织中RhoC、Bmi-1蛋白表达明显高于食管正常黏膜组织(72.5%和30.0%,78.8%和5.0%,P<0.05)。RhoC、Bmi-1的表达与食管浸润深度、淋巴结浸润有关(P均<0.05),但与年龄、民族无关。RhoC和Bmi-1的表达呈正相关(r=0.338,P<0.05)。10年总生存率为20.0%。单因素Kap-lan-Meier生存分析表明,食管癌RhoC、Bmi-1高表达组较低表达组有生存优势,但差异无统计学意义。Cox模型多因素分析表明,RhoC和Bmi-1不是影响生存期的独立因素,而肿瘤浸润深度和转移是影响生存期的独立因素。结论 RhoC、Bmi-1在食管癌组织中表达均较高,其表达水平与食管肿瘤的发生发展有一定的关系,结合临床病理可作为判断预后的指标。Objective To investigate the expression of ras homologue (RhoC) and B-ceil-specific Moloney murine leukemia virus integration site 1 protein(Bmi-1) in esophageal squamous cell carcinoma (ESCC) and to analyze their prognostic value. Methods The expression of RhoC and Bmi-1 Protein in specimens of esophageal squamous cancer were detected by immunohistoehemistry. The relationship of RhoC and Bmi-1 expression with ESCC prognosis was evaluated using testing and COX regression model and Kaplan-Meier survival curve analysis. Results The positive rates of RhoC and Bmi-1 expression in esophageal squamous carcinoma were higher than those in normal esophageal tissue (72.5% vs 30.0%, both P 〈0.05; 78.8% vs 5.0%, both P 〈0.05 ). The expression of RhoC and Bmi-1 was correlated with invasion depth, lymph node metastasis and nationality (P〈0.05), but not correlated with Patient's age, tumor size (P 〉0.05). The expression of RhoC was positively correlated with that of Bmi-1 (r = 0. 338, P 〈0.05). The ten-year overall survival rate for all the patients was 20.0% (16/80). Univariate survival analysis suggested that the five-year survival rate for patients with positive expression of RhoC was higher than for patients with that of Bmi-1 though the difference was not significant (P 〈0.05). Conclusion Both RhoC and Bmi-1 were strongly expressed in ESCC. The expression of RhoC and Bmi-1 were positively correlated with ESCC pro gression. Expression of RhoC and Bmi-1 might be used as prognostic parameter in ESCC patients.
