染料木黄酮对未分化鼻咽癌细胞株的增殖抑制作用

摘　　要：目的:探讨染料木黄酮对未分化鼻咽癌细胞的增殖抑制作用。方法:采用MTT法检测染料木黄酮对未分化鼻咽癌细胞株生长的影响,采用流式细胞术检测染料木黄酮作用于未分化鼻咽癌细胞株后细胞周期的变化。结果:染料木黄酮对不同的未分化鼻咽癌细胞株CNE-2和C666-1均具有增殖抑制作用,作用方式呈时间-剂量-效应关系;并可将CNE-2和C666-1细胞阻滞于G2/M期,对CNE-2作用更强;0、30、60、90和120μmol/L染料木黄酮处理CNE-2细胞72h后,G2/M期细胞比例分别为8.75%、34.95%、84.05%、83.80%和79.60%。结论:染料木黄酮对于未分化鼻咽癌细胞株具有一定的增殖抑制作用,本研究为鼻咽癌的治疗提供了新的实验资料。Objective To investigate the cellular responses of genistein treatment in human undifferentiated nasopharyngeal carcinoma（NPC）cells.Methods Effects of genistein on cell proliferation in undifferentiated NPC cell lines,CNE2 and C666-1,were measured using MTT assay and flow cytometric analysis respectively.Results A significant time-dependent and dose-dependent inhibition of cell growth with genistein treatment was found in present study.The cell cycle phase distribution of genistein-treated cells by flow cytometric analysis revealed that genistein caused an accumulation of cells in G2 /M phase in both CNE-2 and C666-1 cells,an effect more profound in CNE-2 cells.Treatment with 0,30,60,90,and 120 μmol /L genistein in CNE-2 cells for 72 h resulted in the changes in the percentage of cells in G2 /M phase from 8.75% to 34.95%,84.05%,83.80%,and 79.60%,respectively.Conclusions Genistein exerts direct anti-proliferative effects on undifferentiated NPC cells in vitro by inducing G2 /M arrest.These findings provide new insights into the potential intervention of NPC with genistein.
