摘 要:目的:提高心肌梗死造模和侧脑室置管后大鼠的存活率以及侧脑室置管的长时间稳固性,为研究大脑内分泌激素对心衰的影响提供基础。方法:健康雄性SD大鼠60只,随机分为甲、乙、丙三组,每组20只。甲组传统方法侧脑室置管,1周后心梗造模;乙组与丙组均先心梗造模,1周后,乙组传统方法侧脑室置管;丙组改良方法侧脑室置管。超声心动图检查均在心梗造模前与之后3周进行。结果:两次造模后大鼠存活率,甲组50.0%,乙组及丙组均85.0%,与甲组相比,乙组及丙组存活率显著提高(P=0.023)。侧脑室置管第10天导管松脱率,甲组62.5%,乙组52.9%,丙组11.8%。丙组松脱率与甲组相比明显下降(P=0.008),与乙组相比显著降低(P=0.010);置管第30天导管松脱率,甲组75.0%,乙组76.5%,丙组17.6%。丙组松脱率与甲组相比显著下降(P=0.005),与乙组相比明显降低(P=0.001);超声结果证明所有存活大鼠心梗造模成功。结论:合理的造模顺序能提高心梗造模及侧脑室置管后大鼠存活率,改良固定方法可明显提高脑室置管稳固性。Objective To improve the survival rate in rats receiving catheter placement in lateral ventricle and the long-term stability of the catheter,so as to provide a basis for studying the effect of brain endocrine hormones on heart failure.Methods 60 healthy male SD rats were randomly divided into three groups:groups A,B,and C(n = 20).Group A received artificial myocardial infarction one week after traditional catheter placement in lateral ventricle.Groups B and C first received myocardial infarction,and one week later,group B traditional catheter placement in lateral ventricle while group C received modified method of catheter placement.Transthoracic echocardiography was performed at baseline and three weeks after myocardial infarction.Results The survival rate of the rats was 50.0% in group A and 85.0% in both groups B and C,and it was significantly improved in groups B and C(P = 0.023)as compared with group A.The rate of catheter loosening was 62.5% in group A,52.9% in group B,and 11.8% in group C ten days after placement.The rate of catheter loosening rate was obviously decreased in group C as compared with group A(P = 0.008)and group B(P = 0.010).Thirty days after placement,the rate of catheter loosening was 75.0% in group A,76.5% in group B,and 17.6% in group C,the rate was markedly reduced in group C in comparison with group A(P = 0.005)and group B(P = 0.001).Echocardiographic examination showed that the model of myocardial infarction model was successfully established in all the survived rats.Conclusions Rational establishment of the model in proper order can improve the survival rate of rats after coronary artery ligation and catheter placement in lateral ventricle.The modified method of catheter placement can significantly improve the long-term stability of the catheter.
