miR-27a、miR-146a在骨性关节炎患者滑膜组织中的表达及临床意义

摘　　要：目的:检测骨性关节炎(OA)患者与非OA患者滑膜中miR-146a和miR-27a的表达情况,并探讨其与该疾病的相关性。方法:应用实时荧光定量聚合酶链反应检测46例滑膜组织(OA患者23例,非OA患者23例)中miR-27a和miR-146a的相对表达量。结果:miR-27a和miR-146a在所有的滑膜组织中均能检测到,根据qRT-PC原理,ΔCt值越高其目的基因在相应组织中的表达水平越低,即OA患者滑膜组织的miR-27a和miR-146a的表达水平较对照组呈低表达趋势(P<0.05)。结论:miR-27a和miR-146a的差异性表达与OA滑膜病理变化有关,可能调控uPA的表达促进OA的发展。Objective To detect the of miR-146a and miR-27a expression in the synovial tissue of knee joint of people with osteoarthritis（OA）and the healthy control people,and to investigate the correlation between miR-146a and miR-27a and OA.Methods The expression of miR-27a and miR-146a in the synovial tissue from 46 persons（23 in OA group,23 in no OA group）was measured by quantitative reverse transcriptionpolymerase chain reaction（RT-PCR）.Results MiR-27a and miR-146a could be detected in all the knee synovial tissue.According to the mechanism for Q-RT-PCR,the higher value of ΔCt was corresponding to the lower level expression of target gene.The expression of miR-27a and miR-146a in synovium of knee joint in osteoarthritis patients was significantly lower than that of the healthy control people（P〈0.05）.Conclusion The lower expression of miR-27a and miR-146a in the synovial tissue may play a crucial role in the pathologic changes of OA,and may modualte uPA expression and promote the development of osteoarthritis.
