摘 要:目的 研究慢性支气管炎患者肺泡巨噬细胞上CD11c和CD14及其胞内TGF β1mRNA的表达情况。方法 对慢性支气管炎缓解期患者和正常对照者各 10例进行局部支气管肺泡灌洗。将支气管肺泡灌洗液中的肺泡巨噬细胞分离、纯化后 ,与藻红蛋白 (phycoerythrin ,PE)和异硫氰酸荧光素 (fluoresceinisothiocyanate,FITC)标记的CD11c和CD14单克隆抗体共同孵育 ,以流式细胞仪分析其CD11c和CD14表达的百分率 ;应用转化生长因子β1(transforminggrowthfactor beta 1,TGF β1)的cDNA探针原位杂交 ,观察肺泡巨噬细胞内TGF β1的表达。结果 (1)慢性支气管炎组肺泡巨噬细胞上CD11c和CD14表达的百分率 (39.17%± 5 .5 6 % ,35 .73 %± 8.0 5 % )均明显高于正常对照组 (16 .2 9%±3 .78% ,15 .2 6 %± 5 .96 % ) (均P <0 .0 0 1) ;(2 )慢性支气管炎患者肺泡巨噬细胞内TGF β1mRNA的表达量明显高于正常对照者 (P <0 .0 1)。结论 慢性支气管炎缓解期患者肺泡巨噬细胞上CD11c和CD14及其胞内TGF β1mRNA的表达增加 。Objective To observe expressions of CD11c、CD14 on alveolar macrophages (AMs) and TGF β1 mRNA in AMs isolated from the patients with chronic bronchitis. Methods AMs in broncho alveolar lavage obtained from 10 patients with chronic bronchitis and 10 normal controls were purified, and treated with PE labeled anti CD11c monoclonal antibody and FITC labeled anti CD14 monoclonal antibody. Expression of CD11c and CD14 on AMs were analysed by flow cytometry; Expression of TGF β1 mRNA in AMs was detected by in situ hybridization. Results High percentages of AMs expressing CD11c and CD14 were detected in patients with chronic bronchitis (39.17%±5.56%, 35.73%±8.05%) compared with normal controls(16.29%±3.78%,15.26%±5.96%)( P <0.001). Expression of TGF β1 mRNA in AMs in patients with chronic bronchitis was significantly increased as compared to normal controls ( P <0.01). Conclusions High expressions of CD11c、CD14 on AMs and TGF β1 mRNA in AMs were detected in patients with chronic bronchitis and may be responsible for chronic inflammation of airways.
