摘 要:【目的】观察多重慢性应激对大鼠下丘脑—垂体—肾上腺素(HPA)轴的影响,探讨穴位埋线对衰老的干预作用。【方法】选用健康3月龄SD雄性大鼠48只,随机分为4组:空白对照组、D-半乳糖组、复合衰老组、复合衰老加埋线组。空白对照组仅给予6周腹腔注射生理盐水。D-半乳糖腹组给予6周腹腔注射D-半乳糖(剂量为120 mg.kg-1.d-1)。复合衰老组给予6周腹腔注射D-半乳糖(剂量为120 mg.kg-1.d-1),同时从第3周开始给予制动应激,应激每天1次,共应激4周;从第3周开始进行噪声刺激,每天1次,每次30 min,共4周;从第3周开始,进行昼夜颠倒的光刺激,共4周。复合衰老加埋线组造模方法同复合衰老组,另外每周埋线1次,共6周。实验结束后断头取血,并取出下丘脑及垂体,检测每组大鼠与HPA轴相关的下丘脑促肾上腺皮质激素释放激素(CRH)、垂体促肾上腺皮质激素(ACTH)、血清皮质醇(CORT)的含量。【结果】①各组大鼠垂体ACTH水平的变化:与空白对照组比较,D-半乳糖组大鼠垂体ACTH水平显著升高(P<0.05);与D-半乳糖组比较,复合衰老组大鼠垂体ACTH水平显著升高(P<0.05);与复合衰老组比较,复合衰老加埋线组大鼠垂体ACTH水平显著降低(P<0.05)。②各组大鼠血清CORT水平的变化:与空白对照组比较,D-半乳糖组大鼠血清CORT水平显著升高,但差异无统计学意义(P>0.05);与D-半乳糖组比较,复合衰老组大鼠血清CORT水平显著升高(P<0.05);与复合衰老组比较,复合衰老加埋线组大鼠血清CORT水平显著降低(P<0.05)。③各组大鼠下丘脑CRH水平的变化:与空白对照组比较,D-半乳糖组大鼠下丘脑CRH水平显著升高(P<0.05);与D-半乳糖组比较,复合衰老组大鼠下丘脑CRH水平显著升高(P<0.05);与复合衰老组比较,复合衰老加埋线组大鼠下丘脑CRH水平显著降低(P<0.05)。【结论】穴位埋线可减缓多重慢性应激下衰老模型大鼠HPA轴的亢进状态,最终延Objective To observe the effect of multiple chronic stress on hypothalamic pituitary adrenocortical (HPA) axis of aging rats, and to explore the intervention of aging with catgut embedding therapy. Methods Forty-eight 3-month-old healthy SD male rats were randomly divided into blank control group, D-galactose group, multiple stress group, and multiple stress plus catgut embedding group. The blank control group wasgiven intraperipheral injection of normal saline for 6 weeks, injection of D-galactose (120 mg.kg-1.d-1) for 6 weeks. and D-galactose group was given intraperipheral Rats in multiple stress group were given the same treatment as D-galactose group, and simutenously were given immobilization once per day, exposure to noise once per day and lasting 30 rain each time, and photostimulation induced by day-night reversion for 4 weeks from week 3. Rats of multiple stress plus catgut embedding group received the same treatment as multiple stress group, and simutenously were given catgut embedding therapy once per week for 6 weeks. After the experiment was ended, the indexes associated with HPA axis such as hypothalamic corticotropin releasing hormone (CRH), hypophyseal adrenocorticotrophic hormone (ACTH) and serum hydrocortisone (CORT) levels were examined. Results The hypothalamic CRH and hypophyseal ACTH levels in the D-galactose group were increased compared with those in the blank control group (P〈0.05) . In the multiple stress group, CRH, ACTH and CORT levels were increased as compared with those in the D-galactose group (P〈0.05) . CRH, ACTH and CORT levels in the multiple stress plus catgut embedding group were decreased compared with those in the multiple stress group (P〈0.05) . Conclusion The catgut embedding therapy can regulate the hyperfunctional HPA axis of the aging rats induced by multiple chronic stress, and then delays aging.
