摘  要:目的考察脂肪来源干细胞-壳聚糖或丝素蛋白(ADSC-CS/SF)杂化体在促进修复大鼠周围神经损伤方面的作用。方法构建ADSC-CS/SF杂化体,修复10mm长的SD大鼠坐骨神经部分损伤,并通过大体观察、足迹分析、组织化学等方法评价ADSC-CS/SF杂化体对大鼠周围神经损伤修复的促进作用。组间坐骨神经指数、再生神经纤维直径比较采用单因素方差分析。结果与空白对照组比较,ADSC-CS/SF杂化体能够较好地促进大鼠坐骨神经再生修复时后肢功能恢复,空白对照组坐骨神经指数(SFI)(-62.2±1.82)比ADSC-CS/SF杂化体SFI(-23.5±1.93)小(F=511.29,P<0.01),空白对照组再生神经纤维平均直径(3.01±0.37)μm比ADSC-CS/SF杂化体平均直径(5.80±0.45)μm小(F=476.05,P<0.01);而大鼠脂肪来源干细胞的SFI(-23.5±1.93)与天然雪旺细胞SFI(-22.3±1.61)比较差异无统计学意义(F=2.13,P>0.05),脂肪来源干细胞再生神经纤维的平均直径(5.80±0.45)μm与天然雪旺细胞再生神经纤维的平均直径(6.38±0.56)μm比较差异无统计学意义(F=6.56,P>0.05)。结论 ADSC-CS/SF杂化体能够促进损伤坐骨神经的再生以及术后功能恢复。Objective To evaluate the therapeutic effect of adipose-derived stem cell- Chitosan/silk fibroin hybrid (ADSC-CS/SF) on the surgically-induced sciatic nerve injury in a rat model. Methods ADSC-CS/SF hybrid was established and used to regenerate rat sciatic nerve of a 10 mm-width surgically-induced sciatic nerve injury. The functional recovery of nerve was assessed by general observation, walking and histology analysis. Results The reconstruction of the injured sciatic nerve was significantly enhanced as indicated by the greater SFI in the ADSC-CS/SF group than that in the control group (-23.5 ± 1.93 vs -62.2 ± 1.82, P 〈 0.01). The mean diameter of regenerated nerve fiber was greater in the ADSC-CS/SF group than that in the control group [(5.80 ± 0.45)μm vs (3.01 ± 0.37)μm, F = 476.05, P 〈 0.01]. The target skeletal muscle was extensively reinnervated. SFI was similar between the ADSC-CS/SF group and the Schawan cell group (-23.5 ± 1.93 vs -22.3 ± 1.61, F = 2.13, P 〉 0.05). Also, the mean diameter of regenerated nerve fiber was similar between the ADSC-CS/SF group and the Schawan cell group [(5.80 ± 0.45)μm vs (6.38 ± 0.56)μm, F = 6.56, P 〉 0.05]. Conclusions ADSCSC/SF hybrid implantation significantly improves axonal regeneration and functional recovery following surgically-induced sciatic nerve injuries, which suggest that ADSC transplantation can serve as a new potential therapeutic method for sciatic nerve injury.
