不同时机介入"醒脑开窍"针刺法对脑缺血再灌注大鼠SOD活性和MDA含量的影响

摘　　要：目的:观察不同时机介入"醒脑开窍"针刺法治疗对脑缺血再灌注模型大鼠超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。方法:将健康成年雄性SD大鼠按随机对照原则分为假手术组、模型组、3 h针刺组、6 h针刺组、药物组,采用线栓法制备脑缺血再灌注大鼠模型,模型制备成功后,针刺组分别于造模成功后3 h、6 h予以"醒脑开窍"针法治疗,药物组于在造模成功5 h后予香丹注射液腹腔内注射,每日1次,治疗3次。治疗72 h后采集标本检测血清及脑组织中SOD的活性和MDA的含量。结果:与模型组比较,5 h药物组血清SOD活性明显降低(P<0.01),3 h针刺组血清MDA的含量明显升高(P<0.01);6 h针刺组血清SOD活性高于3 h针刺组和5 h药物组(P<0.01),而6 h针刺组血清MDA含量、5 h药物组血清SOD活性和MDA含量均显著低于3 h针刺组(P<0.01),6 h针刺组血清SOD活性显著高于5 h药物组(P<0.01)。3 h针刺组脑组织SOD活性和5 h药物组脑组织SOD活性及MDA含量均显著高于模型组(P<0.01);与6 h针刺组比较,3 h针刺组脑组织SOD活性、5 h药物组脑组织内SOD活性和MDA含量均显著升高(P<0.01)。结论:不同时机介入"醒脑开窍"针刺法可不同程度调节血清及脑组织SOD活性水平,增强氧自由基代谢,有效改善脑缺血再灌注引起的缺血性损伤,进而对损伤脑组织起到一定保护作用;且早期介入疗效更佳。Objective： To explore the effects of the Xingnao Kaiqiao acupuncture therapy at different time on SOD and MDA of rats with cerebral ischemia-reperfusion injury.Methods： The male adult SD rats were randomly divided into the sham operation group,the model group,the 3 h acupuncture group,the 6 h acupuncture group,and the XiangDan drug group;The right side cerebral middle artery of rats was blocked by suture to establish the model of cerebral ischemia-reperfusion.After the model was successfully made for three hours and six hours,the acupuncture groups was separately given the ＂Xingnao Kaiqiao＂ acupuncture treatment.After the model was successfully made for five hours,the drug group was intraperitoneally injected with Xiangdan injection.Seventy-two hours later after the treatment,SOD activity and MDA content was examined in the serum and brain tissue.Results： In comparison to the model group,the serum SOD activity of 5 h drug group dramatically decreased（P0.01）,serum MDA content of 3 h acupuncture group significantly increased（P0.01）,serum SOD activity of 6 h acupuncture group was higher than that of 3 h acupuncture group and 5 h drug group（P0.01）,while serum MDA content of 6 h acupuncture group,serum SOD activity and MDA content were lower than those of 3 h acupuncture group（P0.01）,serum SOD activity of 6 h acupuncture group was significantly higher than that of 5 h drug group（P0.01）.SOD activity in brain tissue of 3 h acupuncture group and 5 h drug group as well as MDA content in brain tissue of 5 h drug group were all markedly higher than those of the model group（P0.01）.In comparison of 6 h acupuncture group,SOD activity in brain tissue of 3 h acupuncture group,SOD activity and MDA content in brain tissue of 5 h drug group all significantly increased（P0.01）.Conclusion： Using the Xingnao Kaiqiao acupuncture therapy at different time could modify the level of SOD activity in the serum and brain tissue,strengthens the metabolism of the oxygen free radical,and improves ischemia
