摘  要:[目的]筛选高效苯胺降解菌并研究其降解特性。[方法]通过驯化富集培养,从河南某化工厂活性污泥中分离出1株以苯胺为唯一碳、氮源的高效苯胺降解菌DA-K,并对该菌株进行了生理生化鉴定和生物学降解特性研究。[结果]DA-K菌株呈革兰氏阴性,细胞为杆状,菌落颜色呈灰白色,初步确定为不动细菌属。通过测定,DA-K菌株生长的最适pH为6.0,最适温度为30℃,可在苯胺质量浓度为2 500 mg/L的无机盐培养基上生长良好。DA-K菌株在苯胺浓度为1 000 mg/L,pH 6.0,30℃,180 r/min的条件下振荡培养96 h,苯胺降解率接近80%。[结论]DA-K菌株苯胺降解效率较高,具有实际处理苯胺废水的能力,为构建基因工程菌奠定了基础。[Objective] The research aimed to screen out an efficient aniline-degrading strain and study its degradation characteristics.[Method] By domesticated enrichment culture,an efficient aniline-degrading strain named as DA-K was isolated from activated sludge sample collected from a chemical plant in Henan.DA-K could use aniline as sole carbon and nitrogen sources.The strain was conducted physiological and biochemical experiments,and its biological degradation characteristics were studied.[Result] DA-K strain was G-and rod-shaped,and its colony color was offwhite.It was initially determined as Acinetobacter sp.By measuring,the optimum growth temperature and pH for DA-K were respectively 30 ℃ and 6.0.DA-K strain could grow well in the medium within 2 500 mg/L of aniline.After shaking for 96 h under the conditions of pH 6.0,30 ℃,180 r/min and 1 000 mg/L of aniline,degradation rate of the aniline by DA-K could reach nearly 80%.[Conclusion] DA-K strain had higher aniline degradation efficiency and actual treatment capability of the aniline wastewater,which laid foundation for establishing gene engineering strain.
